纳米中药升白液抗环磷酰胺所致小鼠免疫损伤的研究

摘　　要：目的:探究纳米中药升白液抗环磷酰胺所致小鼠免疫损伤的作用。方法:采用环磷酰胺(CTX)制备Balb/c小鼠免疫损伤模型,检测纳米中药升白液口服给药后不同时间段对环磷酰胺所致小鼠白细胞减少的保护作用。在纳米中药升白液口服给药8 d后,乙醚麻醉处死小鼠,称量胸腺、脾脏并计算其脏器指数;通过组织病理切片观察纳米中药升白液对小鼠肝脏、脾脏病理形态的影响。结果:CTX模型组白细胞数、胸腺及脾脏指数均明显低于正常对照组(P<0.01);纳米中药升白液各剂量组给药8 d后的白细胞数、胸腺及脾脏指数均明显高于CTX模型组(P<0.01或P<0.05)。病理学检查显示,与正常对照组比较,CTX模型组脾组织和肝细胞坏死明显,脾脏和肝脏淤血明显,脾脏多核巨细胞反应明显,脾脏淋巴细胞数量减少;与CTX模型组比较,纳米中药升白液各剂量组脾组织和肝细胞坏死明显减轻,脾脏和肝脏淤血减轻,脾脏多核巨细胞反应明显减少,脾脏淋巴细胞数量回升(P<0.05)。结论:纳米中药升白液对环磷酰胺所致的小鼠免疫损伤具有明显的防护作用。Objective:To study the anti immune injury efftect of nano traditional Chinese medicine Shengbai liquid in mice induced by cytoxan.Methods:Immune injury mouse model was established by CTX.The protection of mouse leukocyte reduction induced by CTX was detected at various time periods after nano traditional Chinese medicine Shengbai liquid oral administration.Mice were killed by ether anesthesia after nano traditional Chinese medicine Shengbai liquid oral administration 8 days.Then the thymuses and spleens of mice were weighed and their indexes were calculated.The influences of nano traditional Chinese medicine Shengbai liquid on pathological morphology of livers and spleens of mice were observed by the pathological sections.Results:Compared with the normal control group,the number of leukocytes and the indexes of livers and spleens of the model group were significantly reduced(P<0.01).Compared with the model group,the number of leukocytes after drug delivery 8 days and the indexes of livers and spleens of the groups of different doses of nano traditional Chinese medicine Shengbai liquid were significantly increased(P<0.01 or P<0.05).Pathological examination showed that the tissue necrosis and congestions of spleens and livers of the model group were obvious,and the multinucleated giant cell reactions of their spleens were obvious,and lymphocytes of their spleens were reduced compared with the normal control group.Pathological examination also showed that the tissue necrosis and congestions of spleens and livers of the groups of different doses of nano traditional Chinese medicine Shengbai liquid were significantly reduced,and the multinucleated giant cell reactions of their spleens were significantly reduced,and lymphocytes of their spleens were significantly increased compared with the model group(P<0.05).Conclusion:Nano traditional Chinese medicine Shengbai liquid could significantly protect mice against immune injury induced by cytoxan.
