摘  要:目的:观察针刺百会穴对阿尔茨海默病(AD)大鼠海马区自噬相关蛋白1(Beclin1)和微管相关蛋白轻链3-Ⅱ(microtubule-associated protein light chain 3-Ⅱ,LC3-Ⅱ)的影响,探讨针刺治疗AD的可能作用机制。方法:将40只雄性SD大鼠随机分为空白组、假手术组、模型组、针刺组,每组10只,空白组常规喂养,不给予任何干预,假手术组在双侧脑室注射人工脑脊液,模型组、针刺组以链脲佐菌素(STZ)双侧脑室注射诱导阿尔茨海默病模型,并确认造模成功。针刺组选取百会穴进行针刺治疗,造模成功后第2天针刺,连续干预28 d。28 d后利用水迷宫检测大鼠学习记忆能力,免疫印迹法(WB)观察脑组织内自噬相关蛋白Beclin-1和LC3-Ⅱ的表达。结果:假手术组逃避潜伏期以及跨越平台次数与空白组比较,差异无统计学意义(P>0.05);与空白组比较,模型组逃避潜伏期延长、跨越平台次数减少,差异有统计学意义(P<0.01),大鼠出现明显的学习记忆障碍;与模型组比较,针刺组逃避潜伏期明显降低、跨越平台次数增加(P<0.05)。假手术组Beclin-1、LC3-Ⅱ蛋白表达与空白组比较,差异均无统计学意义(P>0.05);模型组海马组织中Beclin-11、LC3-Ⅱ蛋白表达均低于空白组(P<0.05);针刺组Beclin-11、LC3-Ⅱ蛋白表达高于模型组(P<0.01)。结论:针刺百会穴可提高阿尔茨海默病大鼠海马区Beclin-1和LC3-Ⅱ的表达,改善大鼠学习记忆能力;针刺百会穴治疗AD的机制之一可能是调控自噬相关蛋白Beclin-1和LC3-Ⅱ的表达。Objective:To observe the effect of acupuncture at Baihui(DU20)on autophagy-related protein 1(Beclin1)and microtubule-associated protein light chain 3-Ⅱ(LC3-Ⅱ)in hippocampus of rats with Alzheimer's disease(AD),and to explore the possible mechanism of acupuncture in treating AD.Methods:Forty male SD rats were randomly divided into blank group,sham-operated group,model group and acupuncture group with ten rats in each group.The blank group was fed routinely without any treatment.The sham-operated group was injected with artificial cerebrospinal fluid into bilateral ventricles,and the other group was injected with streptozotocin(STZ)into bilateral ventricles to induce Alzheimer's disease model,and confirmed that the model is successful.The acupuncture group selected"Baihui(DU20)"for acupuncture treatment,for 28 days.After 28 days,the learning and memory ability of rats was tested by water maze.The expression of autophagy-related proteins Beclin-1 and LC3-Ⅱin brain tissue was observed by western-blot(WB).Results:Water maze results:Compared with the blank group,there was no significant difference in escape latency and times of crossing plateau in sham operation group(P>0.05).The escape latency of the model group was prolonged and the number of crossing platforms was reduced,which was significant compared with the blank group(P<0.01),which showed obvious learning and memory impairment in rats.Compared with the model group,the escape latency of the Baihui group was significantly decrased and the number of crossing platforms increased(P<0.05).WB results:Compared with the blank group,the expressions of Beclin1 and LC3-Ⅱin sham-operated group had no significant difference(P>0.05),while the expressions of Beclin1 and LC3-Ⅱin model group decreased(P<0.05);compared with model group,the expressions of Beclin1 and LC3-Ⅱin acupuncture group increased significantly(P<0.01).Conclusion:One of the mechanisms of acupuncture at Baihui(DU20)for AD may be to improve the symptoms of Alzheimer's disease by regulating the expre
