五种中草药提取物对热应激下秀丽隐杆线虫运动能力的影响

摘　　要：研究五种不同浓度清热解暑药物(金银花、甘草、夏枯草、菊花和芍药)的提取物对热应激下秀丽隐杆线虫三种运动能力的影响作用。用乙醇浸泡提取的方法粗提得到五种中草药的粗提取物,分别配制三种不同浓度(0.31 mg/mL、0.625 mg/mL、1.25 mg/mL)的中草药提取物,用不同浓度的中草药提取物分别处理同步化后L4期的秀丽隐杆线虫,置于20℃下培养10小时后,进行35℃热应激处理,1 h,2 h,3 h后分别对秀丽隐杆线虫的头部摆动频率、身体弯曲频率和基本运动能力这三种运动能力进行观测。结果发现与空白对照组相比这五种中草药均对35℃热应激后的秀丽隐杆线虫的三种运动能力有影响,并对秀丽隐杆线虫均有保护作用,随着热应激时间的增长秀丽隐杆线虫的运动能力呈下降趋势,3 h后秀丽隐杆线虫运动能力大幅度下降,但中草药处理组的运动能力皆强于空白对照组,其中1.25 mg/mL的夏枯草提取物、金银花提取物、甘草提取物以及菊花提取物和0.625 mg/mL的芍药提取物对热应激下的秀丽隐杆线虫保护作用较强。Investigate the effects of different concentrations of Prunella vulgaris,Glycyrrhiza uralensis Fisch,Prunella vulgars L,Dendranthema morifolium and Paeonia lactiflora Pall on the motor ability of C.elegans under the heat stress.Three different concentrations of Chinese herbal medicines were prepared to treat the nematodes in L4 stage.The head swinging frequency,body bending frequency and basic movement ability of each group were measured.The effects of different concentrations on the movement ability of nematodes were analyzed.Three different concentrations of the Chinese herbal extracts all had protective effects on the motor ability of nematodes after heat stress at 35℃,1.25 mg/mL Prunella vulgaris,Glycyrrhiza uralensis Fisch,Prunella vulgars L,Dendranthema morifolium solutions and 0.625 mg/mL Paeonia lactiflora Pall solution was the most obvious protective effect on nematodes under heat stress.
