摘 要:目的评估芪参颗粒对心力衰竭大鼠心脏功能和心肌细胞内钙离子浓度的影响。方法通过左前降支(LAD)结扎手术和高脂饮食喂养诱发心力衰竭大鼠模型,将造模成功的30只存活大鼠随机分为模型组(等体积的生理盐水灌胃)、芪参颗粒组(2.0 g/kg的芪参颗粒灌胃)和美托洛尔组(10.0 mg/kg的美托洛尔灌胃),各10只,10只正常大鼠作为假手术组(等体积的生理盐水灌胃)。观察芪参颗粒对心力衰竭大鼠心脏功能和心肌细胞内钙离子浓度的影响。结果芪参颗粒组的EF和FS均高于模型组,而LVIDd和LVIDs均小于模型组(P<0.05)。HE染色结果表明,芪参颗粒可防止缺血引起的心肌结构受损;天狼星红染色结果表明,芪参颗粒可防止心脏组织中胶原蛋白沉积和纤维化重构。芪参颗粒组的LDL-C、TC和TG水平低于模型组(P<0.05)。芪参颗粒组的心肌细胞内钙离子浓度低于模型组(P<0.05)。结论芪参颗粒治疗可改善心力衰竭模型大鼠的心脏功能、心室重构和血脂代谢。芪参颗粒的心脏保护作用可能与改善心肌钙转运有关。Objective To evaluate the influences of Qishen granules on cardiac function and calcium ion concentration of myocardial cells in heart failure rats.Methods Through the left anterior descending branch(LAD) ligation operation and high-fat diet feeding induced heart failure rat model,30 surviving rats with successful modeling were randomly divided into model group(normal saline with equal volume by gavage),Qishen granules group(Qishen granules at a dose of 2.0 g/kg by gavage) and metoprolol group(metoprolol at a dose of 10.0 mg/kg by gavage),with 10 rats in each group,10 normal rats were selected as sham operation group(normal saline with equal volume by gavage).The influences of Qishen granules on cardiac function and calcium ion concentration of myocardial cells in heart failure rats were observed.Results The EF and FS of the Qishen granules group were higher than those of the model group,while LVIDd and LVIDs were less than those of the model group(P <0.05).The HE staining results showed that Qishen granules could prevent myocardial structural damage caused by ischemia,the sirius red staining results showed that Qishen granules could prevent collagen deposition and fibrosis remodeling in heart tissue.The levels of LDL-C,TC and TG in the Qishen granules group were lower than those in the model group(P<0.05).The concentration of calcium ion in cardiac myocytes of the Qishen granules group was lower than that of the model group(P <0.05).Conclusion The treatment of Qishen granules can improve the cardiac function,ventricular remodeling and lipid metabolism of heart failure model rats.The cardiac protective effect of Qishen granules may be related to the improvement of cardiac calcium transport.
