摘  要:为了评价CYP1A在四氯化碳(CCl4)诱导的建鲤(Cyprinus carpio.Jian)急性肝损伤中的作用,利用精密肝切片(precision-cut liver slices,PCLS)技术,构建了CCl4诱导的建鲤精密肝切片体外损伤模型。研究中通过测定精密肝切片的增值活力、生化指标、肝组织中CYP1A的m RNA表达水平来探讨CYP1A对肝损伤的调节作用。研究结果显示,4~24 mmol·L^-1 CCl4作用4 h后,建鲤精密肝切片培养上清液的谷丙转氨酶(GPT)、谷草转氨酶(GOT)、乳酸脱氢酶(LDH)含量升高,切片匀浆中和丙二醛(MDA)大量产生,超氧化物歧化酶(SOD)和谷胱甘肽(GSH)水平下降,其中,浓度为12mmol·L^-1的CCl4,损伤4h,肝切片的活力维持在65%以上,可以作为造模的条件;75μmol·L^-1α-萘黄酮作用精密肝切片6h后,肝组织中CYP1A的m RNA表达被显著抑制,生化指标显著改善。研究结果表明,CYP1A与CCl4诱导的建鲤肝损伤关系密切,抑制CYP1A的表达可能对肝损伤有保护作用。In order to evaluate the role of CYP1 A in the acute liver injury, a precision liver damage model induced by CCl4 in vitro of Cyprinus carpio.Jian was constructed using precision cut liver slices(PCLS) technology. In this study, the regulation effect of CYP1 A on liver injury was investigated by measuring the activity of liver slices, biochemical enzymes and mRNA expression level of CYP1 A in liver tissue of precision liver sections. The results showed that the levels of glutamate transaminase(GOT),glutamate transaminase(GPT), lactate and dehydrogenase(LDH) in the supernatants were significantly increased, superoxide dismutase(SOD) and glutathione(GSH) levels in liver tissue homogenates were declined with significant difference comparing with the control group and promote malondialdehyde(MDA) synthesis was promoted after treamentment with 4-24 mmol·L^-1 CCl4 for 4 h. Among them, 4 h after treatment with 12 mmol·L^-1 CCl4, the activity of liver slices was maintained at more than 65%. Treatment with 12 mmol·L^-1 CCl4 on PCLS for 4 h could be used as a condition for modeling. The mRNA expression of CYP1 A in liver tissues was significantly inhibited and the biochemical indexes were significantly improved after 6 h of precision liver slices treated with 75 μmol·L^-1 alpha-naphthalene flavonoids. The results showed that CYP1 A was closely related to liver injury induced by CCl4, and inhibiting the expression of CYP1 A might have a protective effect on liver injury.
