摘　　要：PI3K/AKT/mTOR信号通路涉及细胞的生长、增殖、代谢等重要的细胞生理过程,该通路的异常激活与肿瘤的发生与转移密切相关。PI3K(磷脂酰肌醇3-激酶)是PI3K/AKT/mTOR信号通路的起始节点,其突变并异常激活已在多种癌症中被证实,在肿瘤细胞的增殖、迁移、侵袭、血管生成等过程中扮演着重要角色,因此PI3K成为肿瘤靶向治疗的热门靶点。迄今为止靶向PI3K抑制剂已有4种批准上市,另有数十种处于临床试验阶段。本文综述了PI3K抑制剂单独和联合用药抗肿瘤的研究进展,以期对PI3K抑制剂的进一步开发与应用提供参考。The PI3K/AKT/mTOR signaling pathway is involved in cell growth,proliferation,metabolism and other important cellular physiological processes.And the abnormal activation of this pathway is closely related to the occurrence and metastasis of tumor.PI3K(phospholipid inositol 3-kinase)is the starting node of PI3K/AKT/mTOR signaling pathway.Its mutation and abnormal activation have been confirmed in many types of cancers,which play an important role in tumor cell proliferation,migration,invasion,angiogenesis and other processes.Therefore,PI3K has become a hot target in targeted tumor therapy.So far,four PI3K inhibitors have been approved,and dozens of others are in clinical trials.In this paper,the research progress of PI3K inhibitors alone or in combination with other drugs for tumor therapy is reviewed,in order to provide information for further development and application of PI3K inhibitors.
