摘 要:利用60对分布于玉米10条染色体上的引物,对新科910及其父母本进行SSR分析。从中筛选出15对条带清晰、重复性好的引物,用于构建新科910指纹图谱,并将其数字化,计算出出现相同指纹图谱的概率为4.44×10^-16,概率极低,说明以这15对引物构建的新科910指纹图谱鉴定其真实性是可行的。这15对引物分别是phi96100y1、umc2105k3、bnlg2291k4、umc1705w1、bnlg2305k4、bnlg161k8、phi299852y2、umc2160k3、umc1125y3、bnlg240k1、umc2084w2、umc1231k4、phi041y6、umc1432y6、umc2163w3。其中,umc1705w1、bnlg2305k4、umc1432y6、umc2084w2、phi041y6、umc2163w3这6对引物可用来鉴定新科910纯度。任选1对引物umc2084w2用来检测新科910的纯度,结果为97.30%,与预期结果(97%)基本相符。因此,用这6对引物来鉴定新科910纯度是可行的、结果是可信的。We used 60 pairs of primers which distribute on the 10 chromosomes of maize for the SSR analysis of the maize hybrid Xinke 910 and its parents.15 pairs of primers were selected which were clear and good repeatability of their electrophoresis strip to construct the fingerprint of Xinke 910.And we digitized the fingerprint of Xinke 910 to calculate the probability of the same fingerprint,which was 4.44×10^-16 and this probability was very low.This explains that it is feasible to identify the authenticity of Xinke910 by the fingerprint which is constructed by the 15 pairs of primers.The 15 pairs of primers were phi96100y1,umc2105k3,bnlg2291k4,umc1705w1,bnlg2305k4,bnlg161k8,phi299852y2,umc2160k3,umc1125y3,bnlg240k1,umc2084w2,umc1231k4,phi041y6,umc1432y6,umc2163w3.There were 6 pairs of primers can be used to identify the purity of Xinke 910 which were umc1705w1,bnlg2305k4,umc1432y6,umc2084w2,phi041y6 and umc2163w3.Among them,we selected 1 pair of primer arbitrary,which was umc2084w2 to identify the purity of Xinke 910,the result was 97.30%,which fit the expected results(97%)basically.Therefore,it was feasible and the results were credible to identify the purity of Xinke 910 by the 6 pairs of primers.
