摘  要:目的通过研究延胡索总生物碱(TAC)对犬急性心肌缺血的保护作用,探讨TAC对心肌缺血的作用机制。方法取比格犬,分为模型组、TAC高、中、低剂量组和阳性组。采用结扎犬左冠状动脉前降支制备心肌缺血模型,于结扎前、后及给药后取血,测量血压和心外膜电图,探究TAC对心肌缺血药效学作用。检测抗氧化反应、血管内皮活性物质等指标探究机制。结果TAC可抑制心肌梗死面积和范围。与模型组比较,TAC各组对血压、心率及血液相关指标具有明显药效学作用。结论TAC可抑制血压、升高心率,降低心肌缺血程度和梗死面积。作用机制是多靶点的,可增强心肌抗氧化能力,改善血液流变性,防止血液高凝和血栓形成。Objective To study the protective effect of the total alkaloids of Corydalis yanhusuo(TAC)on acute myocardial ischemia in experimental canines,and to explore the mechanism of the effective parts of Corydalis yanhusuo on myocardial ischemia.Methods Beagle dogs are divided into 5 groups:model group,high,middle and low dose group of TAC,and positive group.Myocardial ischemia model is established by ligating the left anterior descending coronary artery of the dog.Blood is taken before,after coronary ligation,and after administration.The blood pressure and epicardial electrocardiograms of experimental animals are mapped to study the pharmacodynamic effects of TAC on myocardial ischemia.The pharmacodynamic effects of TAC on myocardial ischemia are studied by the changes of blood pressure,heart rate,ischemia degree and range,and myocardial infarction area.The mechanism is investigated by detecting indexes of anti-oxidation reaction and vascular endothelial active substances.Results TAC can inhibit the increase of myocardial infarction area and range.Compared with the model group,the TAC groups had significant pharmacodynamic effects on blood pressure,heart rate and blood related indexes.Conclusion TAC can inhibit the increase of blood pressure and heart rate,effectively reduce the degree of myocardial ischemia and the area of ischemic myocardial infarction.The protective mechanism of TAC on myocardium is multi-target,which can improve the antioxidant capacity of myocardium and improve the blood rheology,prevent the formation of blood hypercoagulation and thrombosis.
