摘 要:目的探索中药有效单体穿心莲内酯与不同种类聚合物沉淀抑制剂的相容性及对沉淀抑制剂玻璃化转变温度的影响。方法选择六种聚合物沉淀抑制剂,以溶剂法制备穿心莲内酯与沉淀抑制剂的固态溶液,利用差示扫描量热法进行分析,测定固态溶液的玻璃化转变温度。结果Kollidon VA64、Soluplus及Eudragit L100-55与穿心莲内酯具有良好的相容性,含有不同比例药物的固态溶液,受药物与聚合物分子间作用力、聚合物抗增塑作用等方面的影响,对三种沉淀抑制剂的玻璃化转变温度产生了不同趋势的影响。结论Kollidon VA64及Soluplus可作为理想的聚合物沉淀抑制剂,用于制备新型穿心莲内酯无定型固体分散体。Objective To explore the miscibility of andrographolide(AG)with different polymer-based precipitation inhibitors(PPIs)and further to evaluate the influences of AG with different ratios on glass transition temperature(Tg)values of PPIs.Methods Six types of PPIs are selected to prepare the solid solution with andrographolide by solvent method.Then the Tg values of the solid solutions are measured by differential scanning calorimetry(DSC).Results AG has good miscibility with Kollidon VA64,Soluplus and Eudragit L100-55.Different ratios of the drugs in the solid solution could have varying degrees of influences on the Tg values of the three types of PPIs through anti-plasticizing effect of ploymers and/or the molecular force between PPIs and drug.Conclusion Kollidon VA64 and Soluplus can be used as ideal PPIs to prepare novel AG amorphous solid dispersion.
