摘  要:目的改进转氨基作用与氨基酸纸层析方式和方法,提高实验效率。方法在新的转氨基反应容器和点样方式下,比较缓冲液类型、转氨基时间、肝匀浆浓度及保存时间、磷酸吡哆醛用量对转氨基的影响。结果适当缩短反应时间、适当稀释和保存肝匀浆对转氨基影响较小;缓冲液类型和磷酸吡哆醛不影响转氨基。结论本研究改进了转氨基反应条件和点样方式,提高了实验效率,改善了实验结果。Objective To improve the pattern and method of transamination and paper chromatography for high experiment efficiency. Methods Compare the effects of substrate’s pH,type of buffer,time of reaction,concentration and preservation time of liver homogenate and the amount of pyridoxal phosphate on transamination under new reaction container and sampling way.Results Shortening the time of reaction,diluting and storing liver homogenate moderately had slight impact on transamination,but the type of buffer and pyridoxal phosphate had no effect on transamination.Conclusion The conditions of transamination and sampling way are improved,and the experiment efficiency as well as results are perfected greatly.
