摘 要:燕麦籽粒皮裸性是燕麦籽粒带壳与籽粒不带壳(裸粒)的一对相对性状,反映了籽粒脱离谷壳的难易程度,研究该性状对作物育种、起源驯化具有重要作用。以二倍体皮、裸燕麦为材料,采用电子克隆的方法,克隆到2个AP2/ERF转录因子基因AsNud1和AsNud2,对这2个基因的结构分析发现,AsNud1和AsNud2为旁系同源基因;二者与HvNud都具有很高的相似性,其中,AsNud2与HvNud的相似性更高;皮燕麦的AsNud1和AsNud2编码的氨基酸有提前终止现象,而裸燕麦中的这2个基因则没有此现象。对AsNud1和AsNud2表达特性分析发现,AsNud1和AsNud2在皮、裸燕麦的花器官中都有表达,但表达模式不同,其中,AsNud1在CIav2921中的表达量低于在CIav9008中的表达量,但二者差异没有达到显著水平;而AsNud2在CIav2921中的表达量显著高于在CIav9008中的表达量。因此推测,AsNud可能参与调控燕麦籽粒麸皮的发育。Oat kernel covering refers to whether the husk wrapped outside the kernel is easy to separate from the kernel after ripening.It is important to study this trait for crop breeding and origin domestication.Two AP2/ERF transcription factor genes,AsNud1 and AsNud2,were cloned from diploid oat by electronic cloning.The structural analysis of the two genes showed that AsNud1 and AsNud2 were homologous genes,these two genes had high similarity with HvNud,among which AsNud2 and HvNud had higher similarity.The amino acids encoded by AsNud1 and AsNud2 in husked oat had early termination,but the two genes in naked oat did not.The expression analysis of AsNud1 and AsNud2 showed that both AsNud1 and AsNud2 were expressed in the flower organs of oats,but the expression patterns were different,the expression of AsNud1 in CIav2921 was lower than that in CIav9008,but the difference was not significant,the expression of AsNud2 in CIav2921 was significantly higher than that in CIav9008.Therefore,it is suggested that gene AsNud may be involved in the regulation of oat kernel development.
