摘 要:为获得新型抗棉铃虫棉花种质材料,利用农杆菌介导法获得新型抗虫基因Vip3A的转化植株,通过卡那霉素、PCR、ELISA鉴定和筛选,并结合室内试验分析这些转基因植株分子特征和抗棉铃虫效果。结果表明,从32个转基因植株中获得22个阳性植株,根据农艺性状表现,筛选出3个转基因株系分别是BV01、BV02、BV03,其对2代棉铃虫的校正死亡率分别达到85.7%、83.2%和86.6%。研究结果可为棉花抗虫育种提供新的抗性种质材料。To obtain new cotton germplasm materials resistant to Helicoverpa armigera,the transgenic plants with Vip3A,a new insect-resistant gene,were developed by Agrobacterium-mediated transformation method.The transgenic plants were identified and screened by kanamycin,PCR and ELISA,and the molecular characteristics and anti bollworm effect of these transgenic plants were analyzed by laboratory experiments.The results showed that 22 positive plants were obtained from 32 transgenic plants.According to the performance of agronomic characters,three transgenic lines were selected,namely BV01,BV02 and BV03.Their corrected mortality to the second generation of cotton bollworm reached 85.7%,83.2%and 86.6%,respectively.The research results can provide new resistant germplasm materials for cotton breeding.
