摘 要:为了探讨钙离子对产溶剂梭菌的影响,通过生物信息学方法对丙酮丁醇梭菌(Clostridium acetobutylicum)ATCC824进行蛋白质组学分析,预测可能存在的钙离子结合蛋白(calcium binding proteins,CaBPs)。利用EF手型(EF-hand)、希腊钥匙(Greek key)、β桶状结构(β-roll)、免疫球蛋白质类似蛋白质(Big-like)结构域基序预测丙酮丁醇梭菌蛋白质组中含有钙离子结合区域的蛋白质,建立本地BLAST数据库,通过GO和KEGG数据库分析所预测到的蛋白质。研究最终预测出228个含有相关位点的蛋白质,利用GO注释在生物过程、细胞组分和代谢过程对蛋白质进行了功能分类。KEGG代谢通路分析表明,该类蛋白质主要参与代谢途径、生物抗性基因合成、次级代谢物生物合成和信号传导等过程。Proteomic analysis was performed on Clostridium acetobutylicum ATCC824 by bioinformatics method to predict the possible presence of calcium ion binding proteins(CaBPs)and to explore the effect of calcium ions on solvent producing Clostridium.Proteins containing Ca2+binding regions in clostridium acetone butanol proteome were predicted by EF-hand loop,Greek key,β-roll,and Big-like motifs.A local BLAST database was established and the predicted proteins were analyzed by GO and KEGG database.Finally,228 proteins containing these sites were predicted and were functionally classified by GO annotation in biological processes,cellular components,and metabolic processes.KEGG metabolic pathway analysis showed that these proteins were mainly involved in metabolic pathways,biosynthesis of antibiotics,biosynthesis of secondary metabolites and signal transduction.
