摘 要:目的:研究皮肤衰老进程中各年龄段大鼠角质形成细胞P53、Sirt1表达差异及松花粉对其表达的干预作用。方法:选用SD大鼠背部皮肤的角质形成细胞为研究对象,比较幼年组、成年组、老年组及松花粉喂养老年组P53、Sirt1表达差异。结果:幼年组、成年组、老年组、松花粉喂养老年组P53表达阳性率(%)分别为(72.99±5.461)%、(58.72±3.515)%、(87.51±5.303)%、(59.63±3.976)%;Sirt1表达阳性率(%)分别为(76.30±3.570)%、(81.81±4.740)%、(61.69±3.904)%、(72.54±5.254)%,差异均有统计学意义(P<0.01)。幼年组P53、Sirt1表达阳性率介于成年组与老年组之间,老年组P53表达阳性率最高,Sirt1表达阳性率最低(P<0.01)。松花粉干预后,松花粉喂养老年组P53表达阳性率降低,Sirt1表达阳性率升高。结论:在皮肤衰老过程中,各年龄段大鼠角质形成细胞P53、Sirt1表达有差异且松花粉有抗皮肤衰老作用。Objective:To compare the P53 and Sirt1 expressions of the rats’keratinocyte in the progress of skin aging and the interventional effect of the pine pollen.Methods:The keratinocyte derived from SD rats’skin of back were employed as the studied cells.The P53 and Sirt1 expressions were compared among rats of infant group,adult group,aged group and the pine pollen-fed aged group.Results:The positive cell rates of the P53 expression of the rats’keratinocyte were(72.99±5.461)%,(58.72±3.515)%,(87.51±5.303)%and(59.63±3.976)%respectively in infant group,adult group,aged group and the pine pollen-fed aged group(P<0.01);The positive cell rates of the Sirt1 expression were(76.30±3.570)%,(81.81±4.740)%,(61.69±3.904)%,(72.54±5.254)%respectively infant group,adult group,aged group and the pine pollen-fed aged group(P<0.01).The positive cell rates of the P53 and Sirt1 expressions of the infant group were between the adult group and the aged group.The aged group showed the highest positive cell rates of the P53 while the lowest Sirt1 expressions(P<0.01).In the pine pollen-fed aged group,the positive cell rates of the P53 expression of decreased significantly,while the positive cell rates of the Sirt1 expression raised significantly.Conclusion:In the process of skin aging,there were differences in the expression of P53 and Sirt1 in keratinocytes of rats of different ages,and pine pollen had the effect of anti-skin aging.
