TFB对过敏性紫癜患儿血清诱导的血管内皮细胞损伤的保护机制

摘　　要：目的探讨鬼针草总黄酮(TFB)对过敏性紫癜(HSP)患儿血清诱导的血管内皮细胞损伤的保护作用及机制。方法用HSP患儿血清对人脐静脉内皮细胞(HUVECs)进行培养,再用TFB干预。实验细胞分为四组:空白组、正常组、HSP组及TFB组。用ELISA法检测细胞上清液中IL-8和TNF-α水平;用RT-PCR和Western blot法检测NF-κB和Fractalkine mRNA和蛋白表达。结果ELISA检测结果显示,与正常组和空白组相比较,HSP组IL-8和TNF-α蛋白表达均明显升高,TFB组IL-8和TNF-α蛋白表达较HSP组明显降低(P<0.05)。RT-PCR和Western blot法检测结果显示,HSP组NF-κB、Fractalkine mRNA和蛋白表达水平明显升高;TFB组NF-κB、Fractalkine mRNA和蛋白表达水平较HSP组明显下降(P<0.05)。结论TFB能抑制NF-κB的高表达,降低HUVECs分泌IL-8和TNF-α,进而缓解血管损伤发生。Objective To investigate the protective effect and mechanism of total flavonoids of bidens bipinnata L(TFB)on vascular endothelial cell injury in children with henoch-schonlein purpura(HSP).Methods Human umbilical vein endothelial cells(HUVECs)were cultured in vitro and intervened with normal and HSP children's serum.Then the HUVECs were divided into blank group,normal group,HSP group and TFB group.Levels of interleukin-8(IL-8)and tumor necrosis factorα(TNF-α)in the supernatant were detected by enzyme-linked immunosorbent assay(ELISA).The mRNA and protein expression of nuclear factor kappa B(NF-κB)and Fractalkine were detected by reverse polymerase chain reaction(RT-PCR)and Western blot in HUVECs.Results ELISA results showed that the expression of IL-8 and TNF-αin HSP group were significantly higher than that in normal control group and blank group(P<0.05),and the level of IL-8 and TNF-αin the TFB group was significantly lower than that in the HSP group(P<0.05);the results of RT-PCR and Western blot showed that the expression levels of NF-κB,Fractalkine mRNA and protein in HSP group were significantly higher than those in normal group and blank group(P<0.05).The expression levels of NF-κB,Fractalkine mRNA and protein in the TFB group were significantly lower than those in the HSP group(P<0.05).Conclusions TFB can inhibit the expression of NF-κB in children with Henoch-Schonlein purpura,reduce the secretion of IL-8 and TNF-αin HUVECs,and then relieve the occurrence of vascular injury.
