摘 要:目的观察6-姜酚对化疗性大鼠异食癖及5-羟色胺(5-HT)水平的影响,初步探讨6-姜酚防治化疗性恶心呕吐(CINV)的相关机制。方法将雄性SD大鼠随机分为空白对照组、6-姜酚正常对照组(50 mg·kg^-1)、昂丹司琼正常对照组(2.6 mg·kg^-1)、模型组、昂丹司琼治疗组(2.6 mg·kg^-1)及6-姜酚高剂量(100 mg·kg^-1)、低剂量(50 mg·kg^-1)治疗组,每组6只;每天分2次灌胃给药。首次给药1 h后,采用腹腔注射顺铂(6 mg·kg^-1)建立大鼠化疗性异食癖模型,以大鼠高岭土摄食量作为其恶心呕吐程度指标。造模24 h后取血、回肠和脑干,采用高效液相电化学法测定各组大鼠外周(血清、回肠)和中枢(延髓)的5-HT及其最终代谢产物5-羟吲哚乙酸(5-HIAA)水平。结果(1)与空白对照组比较,模型组大鼠高岭土摄入量显著升高(P<0.001)。与模型组比较,6-姜酚高、低剂量治疗组大鼠高岭土摄入量显著降低(P<0.01)。(2)与空白对照组比较,模型组大鼠的回肠、延髓、血清5-HT水平均显著升高(P<0.05),模型组大鼠回肠、延髓、血清5-HIAA水平均变化不明显(P>0.05)。与模型组比较,6-姜酚高剂量治疗组大鼠回肠组织的5-HT水平显著降低(P<0.05);6-姜酚高、低剂量治疗组大鼠延髓组织的5-HT水平显著降低(P<0.01)。结论6-姜酚可抑制化疗性大鼠异食癖,具有防治CINV的作用,其机制可能与降低中枢及外周5-HT水平有关。Objective To investigate the mechanism of 6-gingerol on the treatment of chemotherapy-induced nausea and vomiting by observing effects of 6-gingerol on pica and 5-HT level in rats after chemotherapy. Methods SD rats were randomly divided into blank control group, 6-gingerol normal control group, ondansetron normal control group,model group,ondansetron(2.6 mg·kg^-1)treatment group,6-gingerol high-dose(100 mg·kg^-1)and low-dose(50 mg·kg^-1)treatment groups. One hour after the first medication,rats in the model group and the medication groups were injected intraperitoneally with cisplatin(6 mg·kg^-1)to establish chemotherapy-induced pica model. The consumption of kaolin was taken as an indicator of nausea and vomiting. At 24 h after modeling,the levels of 5-HT and its metabolite 5-HIAA in ileum,medulla oblongata and serum were measured by HPLC-ECD. Results(1)Kaolin consumptions in model group were significantly increased(P < 0.001). The high and low dosages of 6-gingerol could significantly inhibit kaolin consumption in cisplatin-treated rats(P < 0.01).(2)In the model group,levels of 5-HT in ileum,medulla oblongata and serum increased significantly(P < 0.05). The levels of 5-HIAA had no significant change(P > 0.05). The high dosage of 6-gingerol could significantly reduce 5-HT level in ileum(P <0.05). The high and low dosages of 6-gingerol could significantly reduce 5-HT level in medulla(P < 0.01).Conclusion 6-gingerol can inhibit chemotherapeutic pica in rats,which suggests that it has the effect of preventing and treating chemotherapeutic nausea and vomiting,and the mechanism may be related to reducing the level of 5-HT in central and peripheral.
