摘 要:【目的】研究广藿香试管苗的根发生过程,为广藿香的组织培养研究奠定基础。【方法】在1/2MS培养基上添加NAA、IAA、IBA 3种激素,设计15种组合用于再生小芽的生根诱导。【结果】3种激素单独或者组合使用均可不同程度诱导广藿香再生芽生根,1/2MS+NAA 1.0 mg/L组,生根率达到81.1%,为最高,但是根从愈伤组织上长出,不利于后期驯化;1/2MS+NAA 0.5 mg/L+IBA 0.5 mg/L组合获得较高生根率,达到80.2%,无愈伤组织影响。试管苗的根呈现2种类型:类型1是光滑的地下丛生根;类型2是密被根毛的气生根转变而来的地下根,即茎基部先长出密被根毛的气生根,之后贴附于基质表面生长,然后再延伸到基质内部生长而成为地下根。【结论】最佳生根培养基为1/2MS+NAA 0.5 mg/L+IBA 0.5 mg/L;广藿香试管苗的根发生有2种类型:光滑的地下丛生根及密被根毛的丛生状气生根转变而来的地下根。【Objective】 To lay a foundation for the micropropagation of Pogostemon cablin, we studied the root organogenesis of test-tube plantlet. 【Methods】 Fifteen combinations of 1/2 MS medium supplementing three auxines(NAA,IAA, and IBA) were designed for root induction from regenerated shoots. 【Results】 The adventitious root was inducted on all mediums with supplemental auxin alone or combination, and the highest rooting percentage(81.1%) was found on 1/2 MS+NAA 1.0 mg/L. However, plantlets with the roots originated from the calli were not suitable for subsequent acclimatization. The suboptimal rooting ratio was 80.2% on 1/2 MS+NAA 0.5 mg/L+IBA 0.5 mg/L without callus interference. Two types of roots were observed: smooth underground cluster roots(type 1) and underground roots transformed from cluster aerial roots closely covered with root hairs(type 2). The cluster aerial roots with dense root hairs grew firstly on the stem base, adhere to the surface of medium, and then fell into the medium as underground roots. 【Conclusion】 The best medium for root induction was 1/2 MS+NAA 0.5 mg/L+IBA 0.5 mg/L. Two types of root were observed: smooth underground cluster root and underground root transformed from cluster aerial root with dense root hairs.
