温经汤对子宫内膜异位症小鼠异位内膜生长及炎症因子水平的影响

摘　　要：目的:观察温经汤对子宫内膜异位症(endometriosis,EMs)小鼠异位内膜生长及炎症因子白细胞介素-1(interleukin-1,IL-1)、IL-6、IL-8、肿瘤坏死因子-α(tumor necrosis factor-α,TNF-α)水平的影响。方法:“腹腔注射子宫内膜法”建立EMs模型,随机分为模型组,温经汤低剂量组、中剂量组、高剂量组及西药对照组各10只,另取未建模5只小鼠作为空白对照组。分别给予温经汤、孕三烯酮和生理盐水灌胃,连续给药21 d后处死小鼠,收集异位病灶并称质量,HE染色法观察异位内膜组织学形态,ELISA法检测血清和腹腔上清液中IL-1、IL-6、IL-8、TNF-α的水平。结果:与模型组病灶质量比较,温经汤中剂量组、高剂量组和西药对照组异位病灶质量显著降低(P<0.05)。异位内膜组织学形态方面,温经汤高剂量组异位内膜较模型组明显萎缩,上皮层厚度降低,血管减少,腔内侧可见细胞脱落。血清和腹腔上清液中炎症因子水平方面,与空白对照组比较,模型组血清和腹腔上清液中IL-1、IL-6、IL-8和TNF-α水平均明显升高(P<0.05),温经汤高剂量组和西药对照组血清IL-1、IL-6、TNF-α水平较模型组显著降低(P<0.05);温经汤高剂量组和西药对照组腹腔上清液中IL-1和TNF-α水平显著低于模型组(P<0.05)。结论:温经汤对EMs模型小鼠异位内膜的生长有抑制作用,同时可以降低炎症因子水平,改善炎症反应。Objective:To observe the effect of Wenjing Decoction on the growth of endometriosis(EMs)and the levels of interleukin-1(IL-1),IL-6,IL-8,tumor necrosis factor-α(TNF-α)in mice with endometriosis(EMs).Methods:The model of EMs was established by the method of"intraperitoneal injection of endometrium".It was randomly divided into model group,Wenjing Decoction low dose group,middle dose group and high dose group,western medicine control group with 10 mice in each group.The mice were killed after 21 days of continuous administration of Wenjing Decoction,gestrinone and normal saline.The ectopic lesions were collected and weighed.The histological morphology of ectopic endometrium was observed by HE staining.The levels of IL-1,IL-6,IL-8 and TNF-αin serum and peritoneal supernatant were detected by ELISA.Results:Compared with the model group,the quality of ectopic focus in Wenjing Decoction middle dose group,high dose group and western medicine control group decreased significantly(P<0.05).In terms of histology and morphology of ectopic endometrium,compared with the model group,the high-dose Wenjing Decoction group significantly shrank the ectopic endometrium,reduced the thickness of epithelial layer,decreased blood vessels,and cell abscission could be seen in the lumen.Compared with the blank control group,the levels of IL-1,IL-6,IL-8 and TNF-αin serum and peritoneal supernatant of the model group increased significantly(P<0.05).The serum levels of IL-1,IL-6 and TNF-αin Wenjing Decoction high dose group and western medicine control group were significantly lower than those in the model group(P<0.05);the levels of IL-1 and TNF-αin the peritoneal supernatant of Wenjing Decoction high dose group and western medicine control group were significantly lower than those in the model group(P<0.05).Conclusion:Wenjing Decoction can inhibit the growth of ectopic endometrium,reduce the level of inflammatory factors and improve the inflammatory response.
