均匀设计结合药效学优选酸枣仁复方提取工艺

摘　　要：目的优选酸枣仁复方的最佳提取工艺条件。方法采用均匀设计法,以乙醇体积分数、溶剂用量及提取时间为优化参数,根据2003年版《保健食品检验与评价技术规范》,开展直接睡眠试验、延长戊巴比妥钠睡眠时间试验及戊巴比妥钠阈下催眠剂量试验,以延长戊巴比妥钠诱导小鼠睡眠时间为评价指标,同时测定干膏得率,优选酸枣仁复方的提取工艺条件,并对优选提取工艺进行验证试验。结果最佳提取工艺为10倍水煎提取2次,每次提取71 min;工艺优化后复方酸枣仁延长戊巴比妥钠诱导小鼠睡眠时间显著长于阳性对照组;干膏得率与药效学指标无明显相关性。结论优选的提取工艺稳定可行、成本低,适合工业生产。Objective To optimize the extraction process of Suanzaoren Formula.Methods According to Technical Standards for Testing&Assessment of Health Food(2013 Edition),the direct sleep-inducing test,prolonging sleeping time induced by pentobarbital and affecting sleeping rate under the threshold hypnogenesis dosage of pentobarbital were conducted,and the prolonging sleeping time induced by pentobarbital was set as the evaluation index.The optimal extraction process of Suanzaoren Formula was studied by uniform design method with respect to ethanol concentration,solvent volume and time,the yield of extraction was determined as well.A demonstration test was conducted by choosing the optimal extraction process.Results The optimal extraction process was extracted twice with 10 times the amount of water,decocted for 71 min per time.The prolonged pentobarbital induced sleeping time of mice in group of formula with optimized extraction process was longer than that of positive control group.The result of correlative analysis showed that there were no significant correlations between the yield of extraction and pharmacodynamic index.Conclusion The optimized extraction process is stable and feasible,which is low cost and suitable for industrial production.
