高效液相色谱法测定复方斑蝥胶囊中4种活性成分含量

摘　　要：目的建立测定复方斑蝥胶囊中马钱苷、特女贞苷、紫丁香苷和甘草苷含量的高效液相色谱(HPLC)法。方法色谱柱为Agilent Zorbax Eclispe Plus C18柱(250 mm×4.6 mm,5μm),柱温为25℃,检测波长为235 nm,进样量为10μL,流动相为乙腈(A)-0.3%磷酸溶液(B),梯度洗脱(0~8 min时5%A,8~15 min时5%~23%A,15~28 min时23%~73%A,28~29 min时73%~5%A,29~35 min时5%A)。外标法定量检测。结果马钱苷、紫丁香苷、甘草苷和特女贞苷质量浓度分别在0.5~50,2.5~250,0.8~80,2.0~200μg/mL范围内与峰面积线性关系良好,加样回收率分别为100.44%,100.62%,100.71%,101.29%,RSD分别为2.92%,2.25%,3.19%,3.05%(n=6)。结论该方法操作简单、速度快、重复性好、检测结果准确,可用于复方斑蝥胶囊的质量控制。Objective To develop a high-performance liquid chromatography(HPLC)method for determination of loganin,nuezhenide,syringin and liquiritin in Compound Banmao Capsules.Methods Agilent Zorbax Eclispe plus C18 column(250 mm×4.6 mm,5μm)was selected for seperation,with column temperature setting at 25℃and detection wavelength at 235 nm;the injection volume was 25μL;acetonitrile was used as mobile phase A and 0.3%phosphoric acid solution as mobile phase B.Gradient elution procedure was as follows:0-8 min 5%A,8-15 min 5%-23%A,15-28min 23%-73%A,28-29 min 73%~5%A,29-35min 5%A.External standard method was selected for quantitative detection.Results The contents of loganin,nuezhenide,syringin and liquiritin showed good linear relationships in the concentration range 0.5-50,2.5-250,0.8-80,2.0-200μg/mL.Sample recovery rates were 100.44%,100.62%,100.71%,101.29%,RSDs were 2.92%,2.25%,3.19%,3.05%(n=6).Test solution was stable within 24 h.Conclusion The method is simple in operation,fast in speed,good in repeatability and accurate in detection results,which can be used for the quality control of Compound Banmao Capsules.
