摘 要:Bt基因具有广谱杀虫性,培育转基因抗虫品种为甘蔗病虫害防治提供了新思路。为了检验Cry2A基因对甘蔗螟虫的防治效果,本研究构建了由玉米Ubi启动子驱动的Cry2A基因高效植物表达载体Cry2A-3300,利用农杆菌介导技术,对甘蔗胚性愈伤组织材料进行遗传转化,经过筛选培养及再生培养共得到46株抗性植株。对抗性植株进行PCR扩增检测,15株呈阳性;进一步利用RT-PCR,表明外源基因Cry2A在转录水平上得到有效表达。室内抗虫鉴定的结果表明Cry2A基因可有效抑制害虫的生长,这将有利于后期进行田间防治。Bt gene has a broad-spectrum insecticidal properties,so developing transgenic insect-resistant varieties provide a new idea for sugarcane pest control.In order to test the control effects of Cry2A gene,we constructed Cry2A-3300,a highly efficient plant expression vector of Cry2A gene driven by maize Ubi promoter.Then Agrobacterium-mediated technology was used for genetic transformation of embryonic callus materials of sugarcane,and 46 resistant plants were obtained through screening and regeneration.These plantlets were detected by PCR and 15 plantlets showed positive.Furthermore,RT-PCR analyses showed that the exogenous gene Cry2A was effectively expressed at the transcription level in these plantlets.The results of laboratory anti-insect identification showed that Cry2A gene could effectively inhibit the growth of pests.
