摘  要:为解决植物缺铁变黄现象,利用嗜铁菌为植物提供铁营养、促进植物生长的研究受到各国学者的广泛关注.该研究利用CAS(铬天青)平板法,从蔬菜根际土壤中分离获得7株嗜铁细菌,铬天青(CAS)法定量检测其中E7和W7菌株产嗜铁素能力较强,分别为71.48%、61.17%,16S rRNA基因序列初步鉴定其分别为苍白杆菌属(Ochrobactrum sp.)、假单胞菌属(Pseudomonas sp.).以E7、W7及其混合菌(E7+W7)为供试菌株,研究对绿豆种子萌发以及土壤中有效铁浓度和嗜铁素活性单位的影响.结果显示,与清水对照相比,菌液处理绿豆种子后能显著增加胚芽、幼根、胚轴长度,以复合菌液(E7+W7)100×稀释液处理效果最佳,胚轴、幼根、胚芽分别显著增加了41.38%、60.65%、55.08%.各菌液处理5天后均能显著增加土壤中亚铁离子浓度,其中以混合菌液处理效果最佳,增加了65.77%,其次是E7和W7,分别增加了57.45%、54.39%;E7+W7和E7处理组在第3天时嗜铁素活性单位分别达到90.71%和85.31%,而W7组在第5天时达到最大值79.07%.To solve the problem of chlorosis,the research on promoting the plant growth by using siderophore-producing bacteria to provide plants with iron nutrition has attracted wide spread attention of scholars all over the world.Seven strains of siderophore-producing bacteria were obtained from the rhizosphere soil of vegetablesby using the chrome azurol S(CAS)plate method.The chrome azurol S(CAS)assay was used for quantitative detection of siderophore,which indicated that E7 and W7 strains have the highest relative levels of siderophore production of 71.48%and 61.17%respectively.They were identified as Ochrobactrum sp.and Pseudomonas sp.respectively by 16 S rRNA gene sequence.E7,W7 and their mixed bacteria(E7+W7)were used as further strains to study their effect on the germination of mung bean seeds and their effective iron and siderophore contents in soil.The results showed that the length of germ,young root and hypocotyl were significantly increased after the treatment of mungbean seeds compared with the clear water.The compound bacterial solution(E7+W7)which were diluted by 100 times had the best effect,and the hypocotyls,young roots and buds increased significantly by 41.38%,60.65%and 55.08%,respectively.After 5 days of treatment,the concentration of ferrous ions in the soil were significantly increased,among which the mixed bacterial solution had the best treatment effect by increasing of 65.77%,while E7 and W7 increased by 57.45%and 54.39%respectively;In the consortium(E7+W7)and E7 groups,the siderophore content reached 90.71%and 85.31%respectively on the third day,while the W7 group reached the maximum value of 79.07%on the fifth day.
