摘  要:家畜的繁殖性状是畜牧业重要的经济性状之一,FecB基因是公认的多胎主效基因。本试验以布鲁拉美利奴羊与新吉细毛羊杂交一代为研究对像,对不携带FecB基因(++型)和携带FecB基因杂合型(B+型)的1月龄母羊超数排卵后,采用手术法进行活体采集卵泡液,收集并培养卵巢颗粒细胞,利用实时荧光定量PCR分析++型和B+型卵巢颗粒细胞上BMPR-1B、BMP15、GDF9、GnRHR、LHR、FSHR、ESR、AMH共8个基因表达情况。结果显示:++型和B+型卵巢颗粒细胞的BMPR-1B、GnRHR、LHR、FSHR基因mRNA表达水平差异不显著(P>0.05);B+型卵巢颗粒细胞的BMP15、AMH基因mRNA表达水平显著高于++型(P<0.05),GDF9、ESR基因mRNA表达水平极显著低于++型(P<0.01),ESR基因mRNA表达水平显著高低于++型(P<0.05)。本试验对利用羔羊超排技术获得的FecB杂交羊卵巢颗粒细胞繁殖相关基因的表达状况进行了分析,进一步明确了利用羔羊超排技术获得的卵母细胞体外成熟阶段的发育环境,为探讨FecB杂交羊对羔羊超排技术的应答机制、优化卵母细胞体外培养技术提供参考。The reproductive trait of livestock is one most important economic traits in animal husbandry.In this experiment,the hybrid generation of Booroola Merino sheep and Xinji fine wool sheep was used as the research animal.After the superovulation of one-month old ewes without FecB gene(++type)and FecB gene heterozygous type(B+type),the follicular fluid was collected in vivo by surgery,and the ovarian granulosa cells were collected and cultured.The expression of BMPR-1 b,BMP15,GDF9,GnRHR,LHR,FSHR,ESR and AMH in ovarian granulosa cells of++type and B+type were analyzed by real-time fluorescence quantitative PCR.The results showed that:there was no significant difference that BMPR-1 b,GnRHR,LHR and FSHR mRNA expression levels between type++and type B+ovarian granulosa cells(P>0.05).The mRNA expression levels of BMP15 and AMH genes in B+type ovarian granulosa cells were significantly higher than those in the B+type(P<0.05).The mRNA expression level of GDF9 and ESR genes was significantly lower than that of the++type(P<0.01).The mRNA expression level of ESR gene was significantly higher than that of the++type(P<0.05).In this experiment,the expression of genes related to the proliferation of ovarian granulosa cells in FecB hybrid sheep was analyzed,and the hybrid sheep used the lamb superposition technique.The development environment of oocyte in vitro maturation stage obtained by lamb overrow technique was further clarified.This experiment provided a reference for the discussion of the response mechanism of FecB hybrid sheep to lamb overrow technology and the optimization of oocyte culture technology in vitro.
