摘 要:从陡水湖、五指峰、阳明山、赣南师范大学校园和井冈山采集土壤样本103份,采用醋酸钠-抗生素法并结合形态学观察和生物测定的方法,分离得到对致倦库蚊幼虫有毒性的芽孢杆菌菌株52株,有毒株占分离菌株总数的32.70%,其中获得高毒力菌株6株.对6株高毒力菌株进行深入研究,生物测定结果表明6株分离菌株对致倦库蚊幼虫LC 50值在0.69~1.18μL/mL之间,都可以作为潜在的杀蚊幼微生物资源.光学显微镜观察发现伴胞晶体形态以方形晶体为主.通过16S rDNA序列比对鉴定其种属类型,结果表明6株菌均归为苏云金芽孢杆菌(Bacillus thuringiensis).SDS-PAGE结果发现6株菌株均出现了明显的晶体蛋白条带.该研究结果不仅可丰富杀蚊微生物菌种资源库,还对控制蚊媒传染病爆发具有重要的理论和应用价值.103 soil samples were collected from several locations in Doushuihu lake,Wuzhifeng mountain,Yangming mountain,the campus of Gannan Normal University and Jinggangshan mountain,Bacillus spp.strains were separated by sodium acetate-antibiotics combining with morphological observation and bioassay.Among them,52 strains were toxic to the Culex quinquefaseiatus larvae,accounted for 32.70%of the total number of isolated strains.The fermentation broth of six strains showed high toxicity against C.quinquefaseiatus larvae with LC 50 value between 0.69~1.18μL/mL,which could be used as potential mosquito-larvicidal resources.The crystal morphology was mainly square crystal by optical microscope.The PCR amplification of 16S rDNA and sequence analysis revealed that 6 bacteria strains were classified as Bacillus thuringiensis.SDS-PAGE results showed that all the 6 strains found obvious crystal protein bands.The results could enrich mosquito-larvicidal microbial resource strains,and have important theoretical and practical value for controlling the outbreak of mosquito-borne diseases.
