补中助长颗粒对SD鼠GH/IGF-1及OPG/RANK/RANKL的影响

摘　　要：目的:观察补中助长颗粒对SD大鼠GH/IGF-1及OPG/RANK/RANKL系统的影响。方法:50只15日龄SD鼠随机分为正常组、西药组、中药高剂量组、中药中剂量组和中药低剂量组,每组10只。正常组给予生理盐水1 mL/(kg·d)灌胃,西药组给予赖氨肌醇维生素B12口服液1 mL/(kg·d)灌胃,中药高、中、低剂量组分别给予补中助长颗粒4 g/(kg·d)、2 g/(kg·d)、1 g/(kg·d)灌胃。8周后检测各组大鼠血清GH、IGF-1、OPG、RANK、RANKL的水平,以及肋软骨OPG、RANK、RANKL积分光密度值。结果:各组大鼠血清GH、RANK水平、肋软骨RANK、RANKL积分光密度值比较无统计学意义(P>0.05),中药中剂量组血清IGF-1、RANKL水平显著高于其他各组(P<0.05),中药中剂量组血清OPG水平、肋软骨OPG积分光密度值显著高于正常组、中药高剂量组、中药低剂量组(P<0.05或P<0.01)。结论:中剂量补中助长颗粒能够提高SD大鼠血清IGF-1及OPG、RANKL水平,促进肋软骨OPG、RANKL表达,可能是其促生长的作用机制之一。Objective:To observe the effects of Buzhong Zhuzhang Granule on GH/IGF-1 and OPG/RANK/RANKL in SD rats.Methods:50 SD rats(15-day-old)were randomly divided into the normal group,the western medication group,the TCM medication groups(high-dose,medium-dose,low-dose),with 10 rats in each group.The normal group was given Normal Saline(1 mL·kg^-1·d^-1),the western medication group was given Lysinositol Vitamin B12(1 mL·kg^-1·d^-1),and the TCM medication groups were treated with Buzhong Zhuzhang Capsule(4 g·kg^-1·d^-1,2 g·kg^-1·d^-1,1 g·kg^-1·d^-1).The serum levels of GH,IGF-1,OPG,RANK,RANKL,and the IOD values of OPG,RANK,RANKL in costicartilage were measured after 8 weeks of intervention in the groups.Results:There were no statistical differences in serum levels of GH and RANK,and in IOD value of RANK and RANKL among the groups(P>0.05).The serum levels of IGF-1 and RANKL were significantly higher in the medium-dose TCM medication group than those in the other groups(P<0.05).The serum levels of OPG and IOD value of OPG were significantly higher in the medium-dose TCM medication group than those in the normal group,the high-dose TCM medication group and the low-dose TCM medication group(P<0.05,P<0.01).Conclusion:The medium-dose Buzhong Zhuzhang Granule can increase serum levels of IGF-1,OPG and RANKL in SD rats,promote the expressions of OPG and RANKL in costicartilage,which may be one of the mechanisms on promoting growth.
