摘 要:研究草珊瑚多糖在运动性骨骼肌损伤(EIMD)中的干预效果及其可能机制。研究方法:随机将24只雄性SD大鼠分为安静对照组(C)、运动损伤组(M)、自然恢复组(N)、草珊瑚干预组(S)四组。采用连续大强度离心运动(下坡跑四周),跑台坡度-16°,每周运动5 d。实验取材:安静对照组(C)与运动损伤组(M)大鼠在最后一次离心运动结束24 h后,腹主动脉采血和取材;自然恢复组(N)采用自然恢复7 d,草珊瑚干预组(S)灌胃给药草珊瑚粗多糖7d,之后过夜禁食12h后腹主动脉采血和取材。结论:(1)M组骨骼肌细胞结构发生损伤性变化,可见肌节排列紊乱,出现Z线扭曲、Z线波形变化等,N组可见有一定修复与改善,S组则基本修复,肌节排列、Z线结构基本正常;(2)肌酸激酶(CK)、乳酸脱氢酶(LDH)、肿瘤坏死因子(TNF-α)活性升高,同时IL-1β、IL-18蛋白表达在大鼠大强度离心运动后显著性升高,自然修复7 d后仍然保持较高水平;(3)草珊瑚粗多糖在干预7 d后能够有效改善相关酶上午活性,并抑制相关炎性因子的表达水平,显现出草珊瑚能够对运动性骨骼肌微损伤(EIMD)具有良好的改善效果,其可能性机制可能与炎性因子的调控有关。Objective: To study the effect of polysaccharides of Sarcandra glabra on exercise-induced musule damage(EIMD) and its possible mechanism. Research methods: 24 male SD rats were randomly divided into four groups: the quiet control group(C), the exercise damage group(M), the natural recovery group(N), and the Sarcandra glabra intervention group(S). Continuous high-intensity centrifugal movement is used(downhill running around). The slope of the running platform is-16 °, 5 d exercise weekly. Experimental materials: In the quiet control group(C) and the exercise damage group(M), 24 hours after the last eccentric exercise, the blood was collected from the abdominal aorta and the material was collected. The natural recovery group(N) used natural recovery for 7 days, and the Sarcandra glabra intervention group(S) intragastrically administered polysaccharides of Sarcandra glabra for 7 days, and then fasted overnight for 12 hours to collect blood and samples from the abdominal aorta. Conclusions:(1) The structure of skeletal muscle cells in group M was damaged, and the sarcomere arrangement disorder Z-ray distortion, and Z-wave waveform changes, e tc. could be seen. There were some repairs and improvements in group N, and group S is nearly repaired. The sarcomere arrangement and Z-ray structure are basically normal.(2) Activities of creatine kinase(CK), lactate dehydrogenase(LDH), and tumor necrosis factor(TNF-α) Increased. At the same time, the expression of IL-1β and IL-18 protein increased significantly after high-intensity eccentric exercise in rats, and it remained high after 7 days of natural repair.(3) The polysaccharides of Sarcandra glabra can effectively improve the morning activity of related enzymes and inhibit the expression level of related inflammatory factors after intervention for 7 days. It appears that Sarcandra glabra can have a good effect on improving exercise-induced musule damage(EIMD) and its possible mechanism may be related to the regulation of inflammatory factors.
