摘 要:目的:观察长期有氧运动与膳食干预对IR大鼠胰岛miR-7/PDX-1/GLUT2-GCK以及分泌蛋白KIR6.2、SNAP25的影响,以探讨其改善IR的中心机制。方法:7周龄SD雄性大鼠随机分为高脂饮食组和标准饮食组。高脂饮食组进行10周的高脂饲料喂养建立IR模型,随后分为高脂安静组(IRHC组)、高脂运动组(IRHE组)、标准饮食安静组(IRNC组)和标准饮食运动组(IRNE组);标准饮食组分为安静组(NC组)和运动组(NE组)。运动干预大鼠进行跑台运动(20 m/min,60 min/天,5天/周,8周)。于末次运动后24 h,取血测定空腹C肽含量;取胰岛,测定miR-7以及相关蛋白的表达。结果:与NC组相比,IRHC组大鼠体重、空腹C肽均显著升高(P<0.01),miR-7、GLUT2、GCK、KIR6.2表达显著降低(P<0.05~0.01),PDX-1、SNAP25表达显著升高(P<0.01);与IRHC组相比,IRNC组体重、空腹C肽均显著下降(P<0.01),miR-7、GLUT2、GCK、KIR6.2表达显著增加(P<0.05-0.01),PDX-1、SNAP25表达显著降低(P<0.05~0.01);IRHE组体重、空腹C肽显著下降(P<0.01),miR-7、PDX-1、GLUT2、GCK、KIR6.2、SNAP25表达均无显著性差异;IRNE组体重、空腹C肽显著下降(P<0.01),miR-7、GLUT2、GCK、KIR6.2表达显著升高(P<0.05-0.01),PDX-1、SNAP25表达显著下降(P<0.01)。结论:8周有氧运动和/或膳食干预改善大鼠胰岛β细胞功能可能是通过改善miR-7表达,进而影响PDX-1表达,进一步调控GLUT2、GCK信号,并改善胰岛β细胞胞吐功能来实现的。Objective:To observe the effect of aerobic exercise and dietary intervention on miR-7/PDX-1/GLUT2-GCK and exocrine functional proteins KIR6.2 and SNAP25 in the islet of IR rats and elucidate the central mechanism of improving IR by aerobic exercise and dietary intervention. Methods:100 male SD rats aged 7 w were divided into high-fat diet group and standard diet group. Rats in high-fat diet group were fed with high-fat diet for 10 w to establish IR rat model,and IR rats were randomly divided into high-fat sedentary group(IRHC),high-fat exercise group(IRHE),normal diet sedentary group(IRNC)and normal diet exercise group(IRNE). Rats in the exercise intervention group were subjected to exercise on treadmill(20 m/min,0%,60 min/d,5 d/w,8 w). At 24 h after the last exercise,the fasting C-peptide in blood was measured,and expression of miR-7 and related proteins in islet were determined.Results:Compared with NC,body weight and fasting C-peptide in IRHC were significantly increased(P<0.01),the expression of miR-7,GLUT2,GCK and KIR6.2 were significantly decreased(P<0.05~0.01),and the expression of PDX-1 and SNAP25 were increased(P<0.01). Compared with IRHC,weight and fasting C-peptide in IRNC were significantly decreased(P<0.01),the expression of miR-7,GLUT2,GCK and KIR6.2 were significantly increased(P<0.05~0.01),and the expressions of PDX-1 and SNAP25 were significantly decreased(P<0.05~0.01). The weight and fasting C-peptide in the IRHE decreased significantly compared with IRHC(P<0.01)),and there was no significant difference among miR-7,PDX-1,GLUT2,GCK,KIR6.2 and SNAP25.The weight and fasting C-peptide in IRNE decreased significantly compared with IRHC(P<0.01),and the expression of miR-7,GLUT2,GCK,and KIR6.2 were significantly increased(P<0.05~0.01)),and the expression of PDX-1 and SNAP25 were significantly decreased(P<0.01)).
