摘 要:目的:探讨何首乌饮对衰老大鼠肝脏胰岛素信号通路的关键基因IR、IRS-1表达的影响。方法:选用9月龄雄性大鼠分为4组即正常对照组,衰老模型组,何首乌饮组,自然衰老组。饲养到16月龄时取材,qRT-PCR检测肝脏组织IRS1、IRS基因表达的变化;HE染色观察肝脏组织细胞的形态学改变。结果:何首乌饮组和正常对照组的IR、IRS1的基因表达水平明显高于自然衰老组和衰老模型组(P<0.01)。HE染色可见衰老模型组、自然衰老组显示衰老征象,何首乌饮组较衰老模型组、自然衰老组衰老征象表现相对较轻。结论:调控胰岛素信号通路中关键基因的表达水平,达到减缓大鼠肝脏组织衰老的作用。Objective:To observe the effect of Heshouwuyin on the expression of insulin pathway IR and IRS-1 genes in liver tissue of aging rats.Methods:Sixty male rats of clean grade 9 months were selected to divide into 4 groups:normal control group,aging model group,natural aging group,and Heshouwuyin group.After feeding 16 months,all groups were taken materials and reserved.Real-time fluorescence quantitative PCR was used to detect the expression of IRS1 and IR genes in liver tissues,HE staining were used to observe the aging of liver tissue.Results:The expression of IR and IRS1 in the group of Heshouwuyin and control group were significantly higher than that of the natural aging group and the model group(P<0.01).Through HE staining,the natural aging group and model group showed aging signs,the Heshouwuyin and young group’s aging signs in relatively lower level than the model group.Conclusions:Heshouwuyin could delay the aging of rat liver by increasing the expression of IRS1 and IR,the key gene of insulin pathway in the liver of aging rats.
