摘 要:目的:探讨丹红滴丸对心肌缺血再灌注损伤大鼠的保护作用及其对炎症的抑制作用。方法:24只雄性大鼠随机分为假手术组、模型组、丹红滴丸组、丹七片组。左冠状动脉前降支结扎45 min/松开恢复血流3 h的方法制备心肌缺血再灌注损伤模型,假手术组只穿线不结扎,给药组缺血即刻十二指肠给药。缺血再灌结束后,摘取心脏,腹主动脉采血,收集血清。N-BT染色计算心肌缺血梗塞范围;HE染色观察缺血再灌心肌组织病理改变;酶联免疫法检测血清中IL-6、IL-1β、TNF-α、ICAM-1水平;免疫组化法检测心肌组织NF-κB蛋白表达水平。结果:与假手术组比较,模型组大鼠心肌组织可见炎性细胞浸润,心肌梗塞区占心肌面积28.89%,血清中IL-6、IL-1β、TNF-α、ICAM-1水平显著升高,NF-κB胞浆表达增多、转核现象明显。与模型组比较,给药组大鼠心肌细胞水肿变性坏死减轻,炎性细胞浸润减少,心肌梗塞面积降低,大鼠血清IL-6、IL-1β、TNF-α、ICAM-1水平降低,心肌NF-κB表达量及核转减少(P<0.05-0.01)。结论:丹红滴丸对心肌缺血再灌损伤大鼠具有保护作用,机制与其抑制缺血再灌注损伤大鼠心肌NF-κB活化,降低体内炎症因子的水平的作用有关。Objective:To explore the protective effect of Danhong dropping pill on myocardial ischemia-reperfusion(MI/R)injury in rats and its inhibitory effect on inflammation.Methods:Twenty-four healthy male Sprague Dawley rats were randomly divided into four groups:Sham-operated group,model group,Danhong dropping pill group and Danqi table group.Myocardial ischemia reperfusion injury model was made by occlusion of the left anterior descending coronary artery for 45 minutes reperfusion in model and drug for 3 hours.The infarct area of myocardial ischemia was calculated by N-BT staining.Myocardium morphology observation was carried out by HE staining.The levels of tumor necrosis factor-α(TNF-α),interleukin-1(IL-1),interleukin-6(IL-6)and intercellular adhesion molecule-1(ICAM-1)were determined by ELISA.The positive expression of NF-κB was detected by immunohistochemistry.Results:Compared with sham-operated group,inflammatory cell infiltration was found in the myocardial tissue of the model group and myocardial infarction area accounts for 28.89%of myocardial area.The levels of IL-6、IL-1β、TNF-αand ICAM-1 in serum were significantly increased(all P<0.05)in MI/R rats.NF-κB increasing expression and nuclear transfer of myocardium can be seen in MI/R rats.Compared with the model group,the myocardial edema,degeneration and necrosis,inflammatory cell infiltration,myocardial infarction area,serum levels of IL-6,IL-1β,TNF-αand ICAM-1,and myocardial NF-κB expression and nuclear transfer were decreased in the drug groups.Conclusion:Danhong dropping pill has protective effect on rats with MI/R injury,and its mechanism is related to its inhibition of NF-κB activation and reduction of inflammatory factor level in vivo.
