摘  要:丹参在治疗冠心病(coronary atherosclerotic heart disease,CHD)的临床实践中占有重要地位,此研究基于整合药理学视角,探索丹参治疗CHD的作用机制,借助中药整合药理学平台中的分析功能,构建丹参治疗CHD疾病的“中药-成分-靶标-通路”多维网络,从多个角度探索丹参治疗CHD潜在的分子机制。在整合药理学平台分析丹参治疗CHD的分子机制研究中发现,丹参对CHD的干预作用可能与多种化合物相关,主要集中于脂溶性的二萜类成分和水溶性的酚酸成分。从关键靶标与作用通路可以看出,丹参治疗CHD的主要作用于COX7C、COXA1、SDHC、ATP1A1、COX5A基因等,涉及氧化磷酸化、帕金森病、能量代谢、循环系统、内分泌系统、甲状腺激素信号通路、幽门螺杆菌感染中的上皮细胞信号转导、柠檬酸循环等信号通路。研究为丹参治疗CHD的临床应用提供了理论基础及研究方向。Danshen(Salvia miltiorrhiza)plays an important role in the clinical practice of coronary heart disease(CHD).Based on the integrated pharmacology,this article investigated the mechanism of Danshen(Salvia miltiorrhiza)in the treatment for CHD.With the aid of analytical functions from the integrated pharmacology platform,a multidimensional network for Danshen(Salvia miltiorrhiza)treating CHD disease of"Chinese medicine-ingredients-target-pathway"is constructed to study the potential molecular mechanism of Danshen(Salvia miltiorrhiza)in the treatment of CHD from the multiple perspectives.The results showed that the intervention of Danshen(Salvia miltiorrhiza)on CHD may be related to a variety of compounds,mainly concentrated in the composition of fat-soluble diterpenoids and water-soluble phenolic acid.From key targets and pathways,Danshen(Salvia miltiorrhiza)in the treatment of CHD mainly influences the genes of COX7C,COXA1,SDHC,ATP1A1,COX5A,containing oxidative phosphorylation,Parkinson's disease,energy metabolism,circulatory system,endocrine system,thyroid hormone signal pathways,epithelial cell signal transduction in Helicobacter pylori infection and citrate cycle signaling pathways.The present work provides a theoretical basis and research direction for clinical application of Danshen(Salvia miltiorrhiza)in the treatment of CHD.
