摘 要:目的探讨黄芪、莪术配伍联合顺铂(DDP)影响肿瘤新生血管生成的作用机制。方法构建人肝癌裸鼠原位移植瘤模型。成瘤后,将其随机分为8组:模型对照组、阳性对照组(DDP)、黄芪莪术配伍高剂量组(H)、黄芪莪术配伍中剂量组(M)、黄芪莪术配伍低剂量组(L)、黄芪莪术配伍高剂量+顺铂组(H+DDP)、黄芪莪术配伍中剂量+顺铂组(M+DDP)、黄芪莪术配伍低剂量+顺铂组(L+DDP),每组8只,给药15 d,Western Blot及Real-time RT-PCR检测肿瘤组织中TF、HGF、FVⅡ蛋白及mRNA的表达。结果与模型对照组相比,H、M、H+DDP、M+DDP组TF、HGF、FVⅡ蛋白及mRNA表达均显著下降(P<0.01),并与剂量有一定的正相关性。与DDP组相比,H+DDP组TF、HGF、FVⅡ蛋白及mRNA表达降低(P<0.01)。结论黄芪、莪术配伍可能是通过下调TF、HGF、FVⅡ蛋白及mRNA表达对肿瘤新生血管生成产生影响。Objective To study themechanisms of Huangqi(radix astragali) and Ezhu(curcuma) and cisplatin(DDP) on tumorangiogenesis. Method Human hepatocarcinoma cells HepG2 were transplanted into nudemice to form the study model, and the modelmice were divided at random into model group, DDP group, high dosage of Huangqi(radix astragali) and Ezhu(curcuma) group(H group), intermediate dosage of Huangqi(radix astragali) and Ezhu(curcuma) group(M group), low dosage of Huangqi(radix astragali) and Ezhu(curcuma) group(L group), high dosage of Huangqi(radix astragali) and Ezhu(curcuma) and cisplatin group(H+DDP group), intermediate dosage of Huangqi(radix astragali) and Ezhu(curcuma) and cisplatin group(M+DDP group), low dosage of Huangqi(radix astragali) and Ezhu(curcuma) and cisplatin group(L+DDP group),8 ineach group. The expressions of TF, HGF, FVⅡ protein and mRNA in tumor tissues were detected by Western Blot and Real-time RT-PCR. Result Compared with the model group, the expressions of TF, HGF, FVⅡ protein and mRNA decreased significantly in H, M,H+DDP, M+DDP groups(P<0.01).It was positively correlated with the dose.Compared with DDP group, the expressions of TF, HGF, FVⅡ protein and mRNA obviously decreased in H+DDP group(P<0.01).Conclusion Compatibility of Huangqi(radix astragali) and Ezhu(curcuma)may affecttumorangiogenesis by down-regulating the expressions of TF, HGF, FVⅡ protein and mRNA.
