摘 要:目的探讨二仙汤对去卵巢大鼠离体心肌缺血再灌注损伤(MIRI)的作用及初步机制。方法36只雌性SD大鼠随机分成假手术组、模型组、戊酸雌二醇组(戊酸雌二醇,8×10^-4g·kg^-1)、二仙汤低剂量组(4 g生药·kg^-1)、二仙汤中剂量组(8 g生药·kg^-1)、二仙汤高剂量组(12 g生药·kg^-1)。除假手术组外,各组动物均摘除卵巢复制更年期综合征大鼠模型,每日给药1次,连续90 d,通过Langdroff心脏灌流系统进行离体心脏灌注,生理信号采集系统记录心率,生化方法检测冠状动脉流出液乳酸脱氢酶(LDH)、谷草转氨酶(AST),心肌组织超氧化物歧化酶(SOD)、丙二醛(MDA)、肌酸激酶(CK-MB)、Ca^2+-ATP酶和Na^+-K^+-ATP酶活力。结果与模型组相比,二仙汤给药组能改善去卵巢大鼠MIRI的心率变化,减少冠状动脉流出液LDH、AST的含量,减少心肌组织MDA的含量,增加心肌组织SOD、Ca^2+-ATP酶和Na^+-K^+-ATP酶活力。结论二仙汤可防治去卵巢大鼠离体MIRI,其机制可能与抗氧化有关。Objective To investigate the effect and mechanism of Erxian Decoction on isolated myocardial ischemia-reperfusion injury(MIRI) in ovariectomized rats. Methods Thirty-six female SD rats were randomly divided into sham group, model group, estradiol valerate group(estradiol valerate, 8×10^-4 g·kg^-1), Erxian Decoction low dose group(4 g drug·kg^-1), medium dose group(8 g drug·kg^-1) and high dose group(12 g drug·kg-1). Except for the sham group, the ovary was removed to replicate the model of climacteric syndrome. The rats in each group were administered once a day for 90 consecutive days. The isolated heart was perfused through Langdroff cardiac perfusion system. The heart rate was recorded by physiological signal acquisition system. The lactate dehydrogenase(LDH), the glutathione transaminase(AST),the superoxide dismutase(SOD), the malondialdehyde(MDA), the creatine kinase(CK-MB), the Ca^2+-ATPase and Na^+-K^+-ATPase activities in myocardial tissue were detected by biochemical methods. Results Compared with the model group, Erxian Decoction could improve the heart rate changes of MIRI in ovariectomized rats, reduce the contents of LDH and AST in coronary effluent, decrease the content of MDA in myocardial tissue, and increase the activities of SOD, Ca^2+-ATPase and Na^+-K^+-ATPase in myocardial tissue. Conclusion Erxian Decoction can reduce the myocardial ischemia-reperfusion injury in isolated ovariectomized rats and its mechanism may be related to antioxidative stress.
