基于中药整合药理学平台的苓桂术甘汤治疗脂质代谢紊乱作用机制

摘　　要：目前中药方剂是临床上治疗疾病的主要方式,但其只是建立在"药物-疾病"或"药物-证型"的研究之上,对于从微观角度探讨方剂中药物化学成分对疾病靶标、信号通路作用的研究甚少。目的通过探究苓桂术甘汤各中药活性成分与脂质代谢紊乱的相关靶标与信号通路之间的关系,从微观角度阐释苓桂术甘汤治疗脂质代谢紊乱疾病潜在的分子机制。方法该研究借助整合药理学平台v1.0网络分析软件,构建苓桂术甘汤治疗脂质代谢紊乱疾病"中药方剂-中药材-化学成分-核心靶标-关键通路"多维网络。通过基因功能和信号通路的富集分析,从多个角度分析预测苓桂术甘汤各中药成分对疾病靶标、信号通路的作用。结果从分析结果中发现,苓桂术甘汤各中药成分可通过神经系统及相关变性疾病、多种氨基酸代谢、能量代谢等信号通路达到对脂质代谢紊乱的治疗作用。结论苓桂术甘汤治疗脂质代谢紊乱可能与人体能量代谢有较大关联。At present, traditional Chinese medicine(TCM) prescription is the main way to treat diseases in clinic, but it is only based on the study of "drug-disease" or "drug-syndrome type". Meanwhile, there is few research about the effects of chemical constituents of traditional Chinese herbal in prescriptions on disease targets and signaling pathways from a microscopic perspective. Objective To explore the relationship between the active components of the herbs in Linggui Zhugan Decoction and the related targets and signaling pathways of lipid metabolism disorders and further explain the potential molecular mechanisms of Linggui Zhugan Decoction in the treatment of lipid metabolism disorder from a microscopic perspective. Method The research was based on integrated pharmacology platform v1.0 network analysis software,and constructed a multidimensional network of "TCM prescription-chemical composition-core target-key pathway" of Linggui Zhugan Decoction in the treatment of lipid metabolic disorders. Through enrichment analysis of gene function and signaling pathway, the effects of different Chinese herbal components of Linggui Zhugan Decoction on disease targets and signaling pathways were analyzed and predicted from multiple perspectives. Result From the analysis results, it was found that the components of TCM herbs in Linggui Zhugan Decoction could treat lipid metabolism disorders through signaling pathways such as neurological diseases and related neurodegeneration, amino acid metabolism and energy metabolism, etc. Conclusion Linggui Zhugan Decoction in the treatment of lipid metabolism disorders may be related to human energy metabolism.
