摘 要:目的探讨中医补肾化痰法对肥胖型多囊卵巢综合征(PCOS)模型大鼠血清及卵巢组织中白细胞介素-1β(IL-1β)、血管内皮生长因子(VEGF)的表达水平。方法选取3周龄雌性SD大鼠60只,采用随机数字表法分为空白组(等量生理盐水灌胃)、模型组(等量生理盐水灌胃)、阳性组(二甲双胍0.043 g/kg)、高剂量组[补肾活血生药3.0 g/(kg·d)灌胃]、中剂量组[补肾活血生药2.0 g/(kg·d)灌胃]和低剂量组[补肾活血生药1.0 g/(kg·d)灌胃]各10只大鼠,除空白组外其余各组均建立肥胖型PCOS模型大鼠,对比各组大鼠连续干预14d后的血清及卵巢组织中IL-1β、VEGF的表达水平。结果模型组的血清IL-1β、VEGF水平显著高于其他组别(P<0.05),空白组和阳性组、中药高、中、低剂量组的IL-1β、VEGF水平比较,差异均不具有统计学意义(P>0.05);与空白组相比,模型组大鼠子宫匀浆液中VEGF浓度明显升高,差异有显著统计学意义(P<0.01);与模型组相比,阳性组大鼠子宫匀浆液中VEGF浓度明显降低,差异有显著统计学意义(P<0.01);中药高剂量组大鼠子宫匀浆液中VEGF浓度降低,差异有统计学意义(P<0.05);其余各组大鼠子宫匀浆液中VEGF浓度无明显差异,不具备统计学意义(P>0.05);与空白组相比,模型组大鼠卵巢匀浆液中IL-1β浓度明显升高,差异有显著统计学意义(P<0.01);与模型组相比,阳性组大鼠卵巢匀浆液中IL-1β浓度明显降低,差异有显著统计学意义(P<0.01);中药高剂量组大鼠卵巢匀浆液中IL-1β浓度降低,差异有显著统计学意义(P<0.05);其余各组大鼠卵巢匀浆液中IL-1β浓度无明显差异,不具备统计学意义(P>0.05)。结论中医补肾化痰法可显著降低肥胖型PCOS模型大鼠血清、子宫及卵巢组织中的IL-1β、VEGF表达水平。Objective To investigate the expression levels of interleukin-1β(IL-1β) and vascular endothelial growth factor(VEGF) in serum and ovary of obese polycystic ovary syndrome(PCOS) model rats treated with kidney-tonifying and phlegm-resolving method. Methods Sixty three-week-old female SD rats were randomly divided into blank group(intragastric administration of equivalent saline), model group(intragastric administration of equivalent saline), positive group(metformin 0.043 g/kg), high-dose group[intragastric administration of 3.0 g/(kg·d) drugs for tonifying kidney and activating blood circulation], medium-dose group[intragastric administration of 2.0 g/(kg·d) for tonifying kidney and activating blood circulation] and low-dose group[intragastric administration of 1.0 g/(kg·d) for tonifying kidney and activating blood circulation]. The obese PCOS model rats were established in all groups except the blank group. The expression levels of IL-1β and VEGF in serum and ovarian tissues were compared after 14 days continuous intervention in each group. Results The levels of serum IL-1β and VEGF in the model group were significantly higher than those in other groups(P<0.05). The levels of IL-1β and VEGF in the blank group and the positive group and the high, medium and low dose groups of Chinese medicine were not statistically different(P>0.05). Compared with the blank group, the concentration of VEGF in the uterus homogenate of the model group was significantly increased and the difference was extremely significant(P<0.01). Compared with the model group, the uterine homogenate of the positive group was significantly decreased(P<0.01). The concentration of VEGF in the uterus homogenate of rats in the high dose group was significantly lower(P<0.05). The concentration of VEGF in the uterus homogenate of the other groups was not obvious. The difference was not statistically significant(P>0.05).
