摘 要:目的对2016年-2018年义乌市输入1型登革热病例进行实验室检测与分子溯源分析。方法采集登革热病例急性期血清标本进行NS1抗原和核酸检测,用RT-PCR法扩增包膜蛋白(Envelope protein,E)基因,测定核苷酸序列并进行系统进化分析。结果6例登革热病例NS1抗原和核酸检测结果均为阳性。6株毒株之间的E基因核苷酸和氨基酸同源性分别为96.6%~99.3%和98.8%~100%,系统进化分析显示,E基因序列均属于登革病毒1型的GⅠ亚型,与东南亚2003年-2016年分离病毒株亲缘关系最近。结论2016年-2018年义乌市输入登革病毒1型的亚型主要为GⅠ亚型,输入来源主要为东南亚国家。Objective To determine the type 1 dengue fever and trace the molecular origin of the pathogen in Yiwu,Zhejiang during 2016-2018.Methods Serum samples from patients in the acute stage were collected to detect NS 1 antigen and nucleic acids,the envelope(E)gene of the virus were amplified by RT-PCR,the amplicons were sequenced and subjected to homologous and phylogenetic analysis.Results Six cases of dengue fever were tested positive for NS1 antigen and nucleic acid.The nucleotide and amino acid homology of the E gene among the 6 strains were within 96.6%-99.3%and 98.8%-100%,respectively.Phylogenetic analysis showed that the E gene sequences belong to the GⅠsubtype of dengue virus type 1,close to the strains isolated in Southeast Asia from 2003 to 2016.Conclusion In 2016-2018,the subtype of dengue virus type 1 imported into Yiwu is mainly GI subtype,mainly from Southeast Asia.
