摘 要:目的探索何首乌在传统炮制方法九蒸九晒过程中游离蒽醌、二苯乙烯苷、没食子酸变化规律,并考察其对大鼠肝脏的影响。方法采用高效液相色谱法测定九蒸九晒何首乌中主要化学成分的含量;采用SD大鼠80只,随机分为8组,即空白对照组、生首乌组、制首乌组及一蒸一晒何首乌组、三蒸三晒何首乌组、五蒸五晒何首乌组、七蒸七晒何首乌组、九蒸九晒何首乌组,空白对照组灌胃生理盐水,其他分别灌胃相应药材水煎液,给药12周,测血清中谷草转氨酶、谷丙转氨酶、碱性磷酸酶含量,取肝脏称重测肝脏指数。结果何首乌中游离蒽醌含量从2.3367 mg·g^-1上升至3.6411 mg·g^-1;二苯乙烯苷的含量从4.9555 mg·g^-1下降至1.0421 mg·g^-1;没食子酸的含量从0.0050 mg·g^-1上升至0.1098 mg·g^-1。与空白组比较,五蒸五晒何首乌组大鼠血清ALT含量明显上升,生首乌组及所有何首乌蒸晒组大鼠血清AST含量明显升高,生首乌组和三、五、七、九何首乌蒸晒组大鼠血清ALP含量明显升高,九蒸九晒何首乌组肝脏重量明显增加。结论何首乌九蒸九晒炮制后,可使游离蒽醌和没食子酸含量明显升高,二苯乙烯苷含量显著下降。对大鼠长时间给予九蒸九晒何首乌水煎液,可造成不同程度肝损伤,随着蒸晒次数的增加,损伤程度先上升后减轻。Objective To determine the changes of free anthraquinone,stilbene glycoside and gallic acid in the traditional process of Polygoni multiflori radix by steaming and sun-drying for 9 times,and to investigate their effect on rat liver.Methods The main chemical constituents in Polygoni multiflori radix were determined by high performance liquid chromatography.Eighty SD rats were randomly divided into 8 groups:a blank control group,aPolygoni multiflori radix group,a Polygoni multiflori radix praeparata group,and a one steaming one sun-drying group,a three steaming three sun-drying group,a five steaming five sun-driying group,a seven steaming seven sun-drying group,and a nine steaming nine sun-drying Polygoni multiflori radix group.The blank control group was given saline,and the others were given decoction of the corresponding medicinal materials for 12 weeks.After the 12 weeks of administration,the contents of ALT,AST and ALP in the serum were measured,and the liver index was measured by liver weighing.Results The content of free anthraquinones increased from 2.3367 mg·g^-1 to 3.6411 mg·g^-1,the content of stilbene glycosides decreased from 4.9555 mg·g^-1 to 1.0241 mg·g^-1,and the content of gallic acid increased from 0.0050 mg·g^-1 to 0.1098 mg·g^-1.Compared with the blank group,the content of serum ALT in the rats of five steaming five sun-drying Polygoni multiflori radix group was significantly increased;the content of AST in the serum of rats in the Polygoni multiflori radix group and all the steaming and sun-drying groups was significantly increased;the content of ALP in the serum of rats in the Polygoni multiflori radix group and the three,five,seven,nine steaming and sun-drying Polygoni multiflori radix groups was significantly higher than that in the control group.The weight of liver increased significantly in the nine steaming nine sun-drying Polygoni multiflori radix group.Conclusion After processing the Polygoni multiflori radix,the contents of free anthraquinones and gallic acid are obviously increa
