摘  要:目的探究土鳖虫水提物对小鼠胚胎成骨前体细胞(MC3T3-E1)、大鼠肾上腺髓质嗜铬瘤细胞(PC12)、人脐静脉内皮细胞(HUVEC)增殖及H2O2损伤修复的影响。方法采用煎煮法制备土鳖虫水提物。以MC3T3-E1、PC12和HUVEC为研究对象,利用MTT法检测土鳖虫水提物对3种细胞的增殖活性的影响,以计数法检测MC3T3-E1倍增时间变化。利用H2O2建立损伤MC3T3-E1、PC12、HUVEC模型,以细胞增殖活力恢复情况评价土鳖虫水提物对H2O2损伤MC3T3-E1、PC12、HUVEC的修复作用。结果土鳖虫水提物作用于MC3T3-E1、PC12、HUVEC后具有显著的促增殖作用,最大促增殖率分别为150.65%、189.00%、108.07%(P均<0.001)。加药组MC3T3-E1倍增时间比阴性对照组细胞倍增时间缩短了1.62 h,也说明土鳖虫水提物对MC3T3-E1具有显著的促增殖作用。MC3T3-E1、PC12、HUVEC经H2O2损伤后,细胞活力分别降为53.87%、56.17%、53.66%(P均<0.001),加药后3种细胞活力最高恢复至94.61%、121.00%、121.72%(P均<0.001),证明土鳖虫水提物对H2O2损伤的MC3T3-E1、PC12、HUVEC具有修复作用。结论土鳖虫水提物对MC3T3-E1、PC12、HUVEC具有显著的促增殖作用,对H2O2损伤的MC3T3-E1、PC12、HUVEC具有修复作用。Objective To determine the effect of water extract of Eupolyphaga sinensis(ESWE)on the proliferation of mouse embryonic osteogenic precursor cells(MC3T3-E1),rat adrenal medulla pheochromocytoma(PC12)and human umbilical vein endothelial cells(HUVEC)and the oxidative damage repairing effect of ESWE on MC3T3-E1,PC12 and HUVEC treated with H2O2.Methods ESWE was prepared by the decocting method.MTT assay was used to measure the proliferative activity of ESWE on the above cells.After being treated with ESWE,the doubling time of MC3T3-E1 was counted.The oxidative damage repairing effect of ESWE on MC3T3-E1,PC12 and HUVEC treated with H2O2 was measured by MTT assay.Results The water extract of ESWE had a significant proliferative effect on MC3T3-E1,PC12 and HUVEC.The maximum proliferative rates of ESWE on the 3 cell lines were 150.65%,189.00%and 108.07%,respectively(P<0.001).The MC3T3-E1 doubling time of the ESWE-treated group was shortened by 1.62 h compared with the negative control group,indicating ESWE’s significant proliferative effect on MC3T3-E1.After MC3T3-E1,PC12,HUVEC were oxidatively damaged by H2O2,and the cell viability decreased to 53.87%,56.17%and 53.66%(P<0.001).While the cell viability recovered to 94.61%,121.00%and 121.72%(P<0.001)respectively after treatment with ESWE.The results showed that ESWE had an oxidative repairing effect on MC3T3-E1,PC12 and HUVEC damaged by H2O2.Conclusion ESWE has a significant proliferative effect on MC3T3-E1,PC12 and HUVEC,and an oxidative repairing effect on MC3T3-E1,PC12 and HUVEC damaged by H2O2.
