摘 要:目的建立大黄HPLC指纹图谱并测定其主要成分含量,并结合化学计量学方法,对不同来源及不同产地的大黄进行一致性评价。方法采用phenomenex C18(4.6 mm×250 mm,5μm)色谱柱,甲醇-0.4%甲酸水作流动相,梯度洗脱,流速1.0 mL·min^-1,检测波长254 nm,柱温30℃。结果12种成分在测定范围内与峰面积线性关系良好(r>0.999),平均加样回收率为96.1%~101.9%。建立的指纹图谱共标定20个共有峰,相似度为0.673~0.950。聚类分析与主成分分析结果一致,栽培掌叶大黄、野生掌叶大黄、药用大黄、唐古特大黄被明显区分开。正交偏最小二乘判别分析得出,已定量的没食子酸、儿茶素、大黄酸-8-O-葡萄糖苷、番泻苷A、大黄酚-8-O-葡萄糖苷和大黄素-8-O-葡萄糖苷对大黄的一致性差异影响较大。结论建立的大黄指纹图谱定量测定方法简便,准确,专属性强,能够通过全指标成分控制药材质量,同时可以实现大黄的一致性评价,区分3种不同来源以及野生与栽培大黄。Objective To establish the HPLC fingerprints of Rhei Radix et Rhizoma,determine the content of its main components,and evaluate the consistency of Rhei Radix et Rhizoma from different origins and different producing areas by chemometrics method.Methods The chromatographic column was phenomenex C18(4.6 mm×250 mm,5μm),the flow phase was methanol-0.4%formic acid water,gradient elution and the flow rate was 1.0 mL·min^-1,the detection wavelength was 254 nm,and the column temperature was 30℃.Results The linear relationship of 12 components was good in the determination range(r>0.999),the average recovery was 96.1%-101.9%.Totally 20 chromatographic peaks were recognized as common peaks in the fingerprints,with similarities at 0.673-0.950.Cluster analysis and principal component analysis were consistent,cultivated Rheum palmatum L,wild Rheum palmatum L,Rheum officinale Baill.and Rheum tanguticum Maxim.ex Balf.were easily differentiated.According to the orthogonal partial least square-discriminate analysis,the quantitative gallic acid,catechin,rhein-8-O-glucosides,chrysophanol-8-O-glucosides,emodin-8-O-glucosides and Senna A had great influence on the consistency difference of Rhei Radix et Rhizoma.Conclusion The established method for the quantitative determination of fingerprint of Rhei Radix et Rhizoma is simple,accurate and specific,and controls the quality of medicinal materials through the whole index components.At the same time,it can evaluate the consistency of Rhei Radix et Rhizoma and differentiate 3 different origins as well as wild and cultivated Rhei Radix et Rhizoma.
