摘 要:目的考察微波真空干燥对胆黄连配方颗粒中间体生物碱类成分的影响。方法采用单因素试验确立微波真空干燥的技术参数,采用HPLC法测定不同中间体中4种生物碱的含量,并与传统常压和减压干燥法进行比较。结果微波真空干燥最佳真空度为-0.08 MPa,微波功率为0.5 kW 15 min+0.3 kW 10 min,清膏相对密度为1.35(50℃);微波真空干燥后的中间体生物碱类成分含量无显著变化,与减压干燥、煎煮液、浓缩液的指纹图谱相似度大于0.999。结论微波真空干燥对胆黄连配方颗粒中间体生物碱类成分无影响,且时间短,干膏质地疏松,该方法稳定可行,适合工业化生产。Objective To determine the effect of microwave vacuum drying on the alkaloid composition in the intermediate of bile processed Rhizoma Coptidis formula particles.Methods The technical parameters of microwave vacuum drying were established by single factor investigation method.The content of 4 alkaloids in different intermediates was determined by HPLC.The quality of the intermediate was compared with that of the traditional vacuum drying.Results The best vacuum degree of microwave vacuum drying was-0.08 MPa,the microwave power was 0.5 kW for 15 min and 0.3 kW for 10 min,and the relative density of the paste was 1.35(50℃).There was no significant change in the content of intermediate alkaloids after microwave vacuum drying,and the similarity of fingerprint was more than 0.999 as compared with vacuum drying,decoction,and concentrated solution.Conclusion Microwave vacuum drying had no effect on the alkaloid composition of the intermediate of bile processed Rhizoma Coptidis formula particles,with short loose dry extract texture time.This method is stable and feasible for industrial production.
