活络止痛丸对风湿痹症模型大鼠的治疗作用及机制研究

摘　　要：目的:观察活络止痛丸对风寒湿痹与风湿热痹模型大鼠的治疗作用,并探究其治疗风湿痹症的作用机制。方法:随机将Wistar大鼠分为风寒湿痹与风湿热痹两大组,每组再分为空白组、模型组、万通筋骨片组、活络止痛丸低剂量和高剂量组,每组各10只。采用弗氏完全佐剂诱导,并通过外界施以"风寒湿"和"风湿热"的致病因素制备风寒湿痹与风湿热痹两类病证结合动物模型,通过检测各组大鼠足肿胀度、关节炎指数、血清白细胞介素-1β(IL-1β)、肿瘤坏死因子-α(TNF-α)、大鼠前列腺素E2(PGE2)、超氧化物歧化酶(SOD)及丙二醛(MDA)水平等,进行本品在治疗风湿功能方面的药效学及作用机制研究。结果:与风寒湿痹与风湿热痹模型组相比,活络止痛丸可显著降低足肿胀度、关节炎指数、血清IL-1β、TNF-α、PGE2、MDA水平,升高血清SOD水平。结论:活络止痛丸对风湿痹症模型大鼠具有治疗作用。Objective:To observe the therapeutic effect of Huoluo Zhitong Pill on wind-cold-damp and wind-damp-heat arthralgia model rats,and explore its mechanism of rheumatism-arthralgia.Methods:Wistar rats were randomized into two major groups,i.e.wind-cold-damp arthralgia group and wind-damp-heat arthralgia group.Each group was further divided into the blank group,model group,Wantongjinggu tablet group,Huoluo Zhitong Pill low-dose and high-dose groups,with 10 in each.Wind-cold-damp arthralgia and wind-damp-heat arthralgia animal models were induced by Freund’s complete adjuvant,and being exposed in wind-cold-damp and wind-damp-heat environment,respectively.To study the pharmacodynamics and mechanism of this product in the treatment of rheumatism,the paw edema,arthritis index,interleukin-1β(IL-1β),tumor necrosis factor-α(TNF-α),prostaglandin E2(PGE2),superoxide dismutase(SOD)and malondialdehyde(MDA)levels in the serum were measured.Results:Compared with the model group of windcold-damp arthralgia group and wind-damp-heat arthralgia,Huoluo Zhitong Pill could significantly reduce the paw edema,arthritis index,IL-1β,TNF-α,PGE2 and MDA levels,and increase SOD levels.Conclusion:Huoluo Zhitong Pill had therapeutic effect on rheumatism-arthralgia model rats.
