摘 要:目的:观察对氯苯丙氨酸(PCPA)致失眠大鼠脑干NG2细胞和NMDA受体mRNA的表达,探讨酸枣仁汤治疗失眠的可能机制。方法:将50只SD大鼠随机分为正常组、对照组、PCPA组、酸枣仁汤组、PCPA+酸枣仁汤组,每组10只,采用腹腔注射PCPA制作失眠大鼠模型,灌胃不同药物后取材,采用RT-PCR检测NG2和NMDA受体mRNA的表达情况。结果:PCPA组NG2细胞、NMDA受体mRNA表达升高,NG2表达量与对照组比较差异有统计学意义(P<0.05),NMDA受体表达量与对照组比较差异有显著统计学意义(P<0.01);与PCPA组比较,PCPA+酸枣仁汤组NG2、NMDA受体mRNA表达下降(P<0.05),差异有统计学意义。正常组和对照组NG2、NMDA受体表达量比较无统计学意义(P>0.05)。结论:PCPA诱导的失眠大鼠NG2细胞和NMDA受体表达升高,而酸枣仁汤可以下调二者表达量,提示酸枣仁汤干预失眠的机制可能与调节NG2细胞和NMDA受体的表达有关。Objective:To observe the expression of NG2 cells and NMDA receptor mRNA in brain stem of rats with insomnia induced by chlorophenylalanine,and to explore the possible mechanism of Suanzaoren Decoction in treating insomnia.Methods:Fifty SD rats were randomly divided into normal group,control group,PCPA group,SZRD group,and PCPA+SZRD group,10 rats in each group.PCPA was injected intraperitoneally to make insomnia models,different drugs were administered to the stomach,and materials were obtained and RT-PCR detection of NG2,NMDAR mRNA expression.Results:The expression of NG2 and NMDAR mRNA in PCPA group increased.The expression of NG2 cells was different from that of control group(P<0.05).The expression of NMDA receptors was significantly different from that of control group(P<0.01).Compared with PCPA group In comparison,the expression of NG2 and NMDAR mRNA in PCPA+SZRD group decreased(P<0.05),and the difference was statistically significant.There was no significant difference between the normal group and the control group(P>0.05).Conclusion:The expression of NG2 cells and NMDA receptors in PCPA insomnia rats increased,and Suanzaoren Decoction could down-regulate the expression of both,suggesting that the mechanism of Suanzaoren Decoction intervention in insomnia may be related to the regulation of NG2 cells and NMDA receptor expression.
