摘  要:目的研究根皮素对对乙酰氨基苯酚诱导的小鼠急性肝损伤的保护作用。方法将昆明种小白鼠随机分为空白组,模型组,低、高剂量阳性对照组和低、高剂量实验组,每组8只。空白组和模型组均灌胃给予等量蒸馏水;低、高剂量阳性对照组分别灌胃给予264.85和529.70 mg·kg-1槲皮素;低、高剂量实验组分别灌胃给予240.00和480.00 mg·kg-1根皮素。6组小鼠每天给药1次,连续7 d。除空白组外,其他5组于第7天,用400 mg·kg-1对乙酰氨基苯酚腹腔注射,诱导小鼠急性肝损伤,24 h后采集血样。用试剂盒法检测谷丙转氨酶(GPT)、谷草转氨酶(GOT)、γ-谷氨酰转肽酶(γ-GT)、碱性磷酸酶(ALP)和总胆红素(TB)。用病理学方法,检查根皮素对小鼠肝损伤的影响。结果模型组和低、高剂量实验组的GPT分别为(354.62±10.82),(265.17±12.89)和(208.06±15.99)unit·L-1,GOT分别为(356.54±36.92),(271.54±28.90)和(227.41±23.11)unit·L-1,γ-GT分别为(265.12±21.85),(173.32±25.98)和(142.62±16.69)unit·L-1,ALP分别为(35.73±1.72),(24.61±2.61)和(19.91±0.56)King unit/100 mL,TB分别为(356.20±32.91),(259.92±23.82)和(220.70±15.75)μmol·L-1。模型组的上述指标与低、高剂量实验组比较,差异均有统计学意义(均P<0.05),且低、高剂量实验组的上述指标相比,差异均有统计学意义(均P<0.05)。低、高剂量实验组的小鼠肝组织病理学病变得到了显著改善。结论根皮素对对乙酰氨基苯酚诱导的小鼠急性肝损伤有显著的保护作用。Objective To study the hepatoprotective effects of phloretin on acetaminophen-induced acute liver damage in mice.Methods Kunming mice were divided into 6 groups, the followings: blank group(water), model group(water), positive control-L and-H groups(264.85 and 529.70 mg·kg-1 quercetin), experimental-L and-H groups(240.00 and 480.00 mg·kg-1 phloretin), eight in each group.Six groups of mice were given drugs once a day for 7 days.Except the blank group, the other 5 groups were injected intraperitoneally with 400 mg·kg-1 acetaminophen on the 7 th day to induce acute liver injury in mice.24 hours later, blood samples were collected.The glutamic pyruvic transaminase(GPT), glutamic oxaloacetic transaminase(GOT), γ-glutamyl transpeptidase(γ-GT), alkaline phosphatase(ALP) and total bilirubin(TB) were detected by kit method.The effect of phloretin on liver injury in mice was examined by pathological method.Results The main indexes of model,experimental-L and-H groups were compared: GPT were(354.62±10.82),(265.17±12.89) and(208.06±15.99) unit · L-1,GOT were(356.54±36.92),(271.54±28.90) and(227.41±23.11) unit · L-1,γ-GT were(265.12±21.85),(173.32±25.98) and(142.62±16.69) unit·L-1,ALP were(35.73±1.72),(24.61±2.61) and(19.91±0.56) King unit/100 mL,TB were(356.20±32.91),(259.92±23.82) and(220.70±15.75) μmol·L-1.There were significant differences in the above-mentioned indexes between model group and experimental-L and-H groups(all P < 0.05),and there were significant differences between experimental-L group and experimental-H group(all P < 0.05).Phloretin produced a significant reduction in the histopathological hepatic lesions.Conclusion Phloretin had obvious hepatoprotective effects on acetaminophen-induced acute liver damage.
