摘 要:目的探究柴胡皂苷A对非酒精性脂肪肝(NAFLD)小鼠的干预效果及作用机制。方法 ICR小鼠以连续饲喂高脂饲料法构建NAFLD小鼠模型,并随机分为模型组及实验组,每组15只;另取15只常规饲养的小鼠作为对照组。对照组和模型组均予以等量0.9%NaCl;实验组按5 mg·kg-1的剂量予以1 mg·mL-1柴胡皂苷A。3组小鼠每天灌胃给药1次,连续给药14 d。用腹腔糖耐量实验检测小鼠胰岛素抵抗,用全自动生化分析仪检测血清脂代谢指标及肝功能指标水平,并进行肝脂肪变性评分。结果治疗后,对照组、模型组和实验组的胰岛素抵抗指数分别为(2.43±1.21),(4.39±0.35)和(3.42±0.31),谷丙转氨酶分别为(46.68±5.64),(79.19±4.22)和(53.89±4.52)U·L-1,谷草转氨酶分别为(25.83±10.36),(183.84±20.39)和(132.17±16.05)U·L-1,三酰甘油分别为(0.41±0.11),(0.76±0.21)和(0.54±0.12)mmol·L-1,总胆固醇分别为(0.88±0.11),(3.64±0.21)和(2.17±0.32)mmol·L-1,肝脂肪变性评分分别为0,(55.16±5.43)和(33.65±3.34)分,模型组的上述指标与对照组和实验组比较,差异均有统计学意义(均P<0.05)。结论柴胡皂苷A可有效抑制NAFLD小鼠肝脂肪变性,恢复肝脂代谢及肝功能,其作用机制可能与改善胰岛素抵抗有关。Objective To investigate the effect and mechanism of saikosaponin A on nonalcoholic fatty liver disease(NAFLD) mice.Methods ICR mice were fed with high-fat diet continuously to construct NAFLD mice model.They were randomly divided into model group and experimental group with 15 mice per group, and another 15 mice were fed routinely as control group.The control group and model group were given the same amount of 0.9% NaCl, while the experimental group was given 5 mg·kg-1 saikosaponin A(1 mg·mL-1).Three groups were given orally once a day for 14 days.Insulin resistance in mice was measured by abdominal glucose tolerance test, serum lipid metabolism and liver function were measured by automatic biochemical analyzer, and liver steatosis was scored.Results The Homeostasis model assessment of insulin resistance in control group, model group and experimental group were(2.43±1.21),(4.39±0.35) and(3.42±0.31) mU·L-1, glutamic pyruvic transaminase were(46.68±5.64),(79.19±4.22) and(53.89±4.52) U · L-1,glutamic oxaloacetic transaminase were(25.83±10.36),(183.84±20.39) and(132.17±16.05) U·L-1, triglyceride were(0.41±0.11),(0.76±0.21) and(0.54±0.12) mmol · L-1,total cholesterol were(0.88±0.11),(3.64±0.21) and(2.17±0.32) mmol·L-1,hepatic steatosis scores were 0,(55.16±5.43) and(33.65±3.34) score.The above-mentioned indexes in the model group were significantly different from those in control group and experimental group(all P < 0.05).Conclusion Saikosaponin A can effectively inhibit liver steatosis and restore liver lipid metabolism and liver function in NAFLD mice.Its mechanism may be related to improve insulin resistance.
