红景天苷对急性呼吸窘迫综合征大鼠肺损伤的保护作用研究

摘　　要：目的研究红景天苷对急性呼吸窘迫综合征(ARDS)大鼠肺损伤保护作用及对肺组织细胞色素氧化酶Ⅱ(COXⅡ)表达的影响。方法将60只SPF级、雄性Wistar大鼠随机分为对照组(n=20)、模型组(n=20)及实验组(n=20)。模型组及实验组大鼠均用气管滴注3.00 mg·kg-1脂多糖(LPS)100μL构建ARDS大鼠模型。造模成功后,实验组大鼠腹腔注射红景天苷注射液8.00 mg·kg-1·d-1,对照组与模型组则注射等量生理盐水,连续干预3 d。干预结束后,收集各组大鼠右肺组织,测肺湿干重比(W/D);以苏木精-伊红(HE)染色观察大鼠肺组织病理学形态变化;用酶联免疫吸附(ELISA)法检测肺组织炎性因子肿瘤坏死因子-α(TNF-α)、白细胞介素-6(IL-6)及干扰素-γ(INF-γ)表达水平;用免疫组化法检测各组大鼠肺组织COXⅡ蛋白阳性细胞百分比。结果对照组、模型组及实验组大鼠肺组织W/D值分别为4.12±0.65,7.64±0.91和5.93±0.96;TNF-α的表达水平分别为(52.61±18.32),(136.00±30.21)和(90.12±21.46)pg·mL-1;IL-6的表达水平分别为(213.00±24.56),(361.00±30.42)和(268.00±21.91)pg·mL-1;INF-γ的表达水平分别为(48.82±4.53),(73.74±6.01)和(60.14±5.22)pg·mL-1;COXⅡ蛋白阳性细胞百分比分别为(3.51±1.25)%,(6.35±2.24)%和(4.63±1.82)%。与对照组相比,模型组和实验组肺组织的W/D比值、炎性因子TNF-α、IL-6及INF-γ和COXⅡ蛋白表达水平均显著升高,且实验组以上指标均显著低于模型组(均P<0.05)。结论红景天苷可减轻大鼠肺组织病变程度,也可有效控制ARDS大鼠炎性反应,下调COXⅡ蛋白表达水平,保护肺组织炎性损伤。Objective To investigate the protective effects of salidroside on lung injury in rats with acute respiratory distress syndrome(ARDS) and its effect on cytochrome oxidase Ⅱ(COXⅡ) expression in lung tissue.Methods Sixty SPF and male Wistar rats were randomly divided into control group(n=20), model group(n=20) and test group(n=20).The ARDS rat models were constructed by tracheal instillation of 3.00 mg·kg-1 lipopolysaccharide(LPS) 100 μL in model group and test group.After successful modeling, rats in test group were injected with 8.00 mg·kg-1·d-1 salidroside injection intraperitoneally, control group and model group were injected with the same amount of normal saline for 3 d.After the intervention, the right lung tissues of each group were collected and the ratio of wet to dry weight(W/D) was measured.Hematoxylin-eosin(HE) staining was used to observe the pathological changes of rat lung tissue.The expression levels of inflammatory factors tumor necrosis factor-α(TNF-α),interleukin-6(IL-6) and interferon-γ(INF-γ) were detected by enzyme-linked immunosorbent assay(ELISA).The percentage of COXⅡ protein positive cells in lung tissue of each group was detected by immunohistochemistry method.Results The W/D ratios of lung tissue in control group,model group and test group were4.12±0.65, 7.64±0.91 and 5.93±0.96, the expression levels of inflammatory factor TNF-α were(52.61±18.32),(136.00±30.21),(90.12±21.46) pg · mL-1,the expression levels of IL-6 were(213.00±24.56),(361.00±30.42),(268.00±21.91) pg · mL-1,the expression levels of INF-γ were(48.82±4.53),(73.74±6.01),(60.14±5.22) pg·mL-1,the percentage of COXⅡ protein positive cells were(3.51±1.25) %,(6.35±2.24) % and(4.63±1.82) %.Compared with control group,the W/D,inflammatory factors TNF-α,IL-6,INF-γ and COXⅡ protein expression levels in the lung tissue of model group and test group significantly increased,and the above indexes were significant lower in test group than those of model group(all P < 0.05).Conclusion Salidroside can
