摘 要:目的研究哮喘模型小鼠肺脏和脾脏组织中记忆T细胞(Tm)不同时期的表达规律以及可能作用机制。方法将84只小鼠随机分为对照组和模型组,每组42只。模型组予以腹腔注射和雾化吸入卵清白蛋白,对照组则予以等量磷酸缓冲盐溶液。于激发前1日开始,每隔2 d处死2只小鼠,直至激发后20 d。收集小鼠脾脏和肺脏,用流式细胞术检测脾脏和肺脏中Tm的水平,实时定量聚合酶链式反应检测T盒子转录因子(T-bet)、维甲酸相关孤儿受体γ亚型(RORγt)mRNA的水平。结果激发后第7天肺脏中CD4^+Tm-(辅助T细胞)Th比例达到最大值为(46.20±2.18)%,第9天脾脏中CD4^+Tm-Th比例为(38.35±1.60)%;第3天肺脏中CD8^+Tm-Th比例达到最大值为(42.29±0.97)%,第7天脾脏中CD8^+Tm-Th比例达到最大值为(31.26±1.04)%。对照组和模型组肺组织中T-bet mRNA的表达量分别为1.00±0.01,0.73±0.07;RORγt mRNA表达量分别为1.02±0.01,2.65±0.33,模型组与对照组比较,差异有统计学意义(均P<0.05)。结论 CD4^+Tm可能参与哮喘小鼠气道的持续炎症反应,CD8^+Tm可能表达于哮喘的急性发作时期,其作用机制可能与Th17、Th1细胞的分化有关。Objective To study the expression of memory T cells(Tm)in lung and spleen of asthmatic mice and the possible mechanism. Methods 84 mice were randomly divided into control group and model group, and 42 mice of each group.The model group was intraperitoneally injected and nebulized by inhalation of ovalbumin to construct a mouse model of asthma, while the control group was given an equal amount of phosphate buffered saline. Two mice were sacrificed every 2 days from the first day before the stimulation until 20 days after stimulation.The spleen and lung of the mice were collected, the levels of Tm in the spleen and lung were detected by flow cytometry;and the level of T-box transcription factor(T-Bet) and retinoic acid-related orphan receptor gamma subtypes(RORγt) mRNA was detected by real-time PCR. Results After stimulation, on the 7 th day, the ratio of CD4^+Tm-Th in the lung reached the maximum(46.20±2.18)%, and the ratio of CD4^+Tm-Th in the spleen on the 9 th day reached a maximum of(38.35±1.60)%;on the third day, the ratio of CD8^+Tm-Th in the lung reached a maximum of(42.29±0.97)%. On the 7 th day, the ratio of CD^8+Tm-Th in the spleen reached a maximum of(31. 26 ± 1. 04) %. The expressions of T-bet mRNA in the lung tissue of the control group and the model group were 1. 00 ± 0. 01 and 0. 73 ± 0. 07,respectively;the expression levels of RORγt mRNA were 1. 02 ± 0. 01 and 2. 65 ± 0. 33,respectively;the difference between the model group and the control group was statistically significant( all P < 0. 05). Conclusions CD^4+Tm may participate in the persistent inflammatory response of the airways and CD8^+Tm may play an important role in the acute episode of asthma. The mechanism was related to the differentiation of Th17 and Th1 cells.
