摘  要:目的观察苏木乙酸乙酯提取物抑制心脏移植模型大鼠心肌细胞凋亡及其作用机制。方法用经改良的Ono术式进行大鼠同种异位心脏移植术,将建模成功的大鼠随机分为实验组、对照组、模型组,每组15只。实验组灌胃苏木乙酸乙酯提取物溶液10 mL·kg^-1(2.35 g·kg^-1生药量),对照组灌胃环孢素A(CsA)10 mg·kg^-1,模型组灌胃等量生理盐水,连续7 d。术后通过腹部触诊判断供心搏动情况,停止搏动作为排斥反应的终点。观察各组大鼠移植心脏生存时间,以苏木精-伊红(HE)染色法观察各组大鼠心肌组织形态学变化,以DNA断裂的原位末端标记法(TUNEL)检测各组大鼠心肌组织细胞凋亡情况,以免疫组化法及蛋白质印迹法检测各组大鼠心肌组织中胱天蛋白酶-3(caspase-3)蛋白表达情况。结果实验组、对照组和模型组大鼠移植心脏平均生存时间分别为(29.53±3.14),(31.25±3.69),(8.65±1.28)d,大鼠Stanford标准分级为2级,2级,4级,大鼠心肌细胞凋亡指数(AI)分别为14.32±3.34,18.96±2.54,46.47±2.37。免疫组化染色可见,实验组、对照组和模型组心肌组织caspase-3蛋白相对表达量分别为(8.41±2.55)%,(10.52±2.34)%,(21.43±2.13)%;蛋白质印迹法检测可见,大鼠心肌组织caspase-3蛋白相对表达量分别为0.96±0.14,2.51±1.03,2.15±0.24,差异均有统计学意义(均P<0.05)。结论苏木乙酸乙酯提取物通过降低大鼠心肌组织caspase-3蛋白表达进而抑制心肌细胞凋亡,减轻心肌损伤,延长大鼠存活期。Objective To investigate the inhibitory effect of ethyl acetate extract of hematoxylin on cardiomyocyte apoptosis in rat heart transplantation model and its mechanism.Methods Rats were randomly divided into test group,control group and model group,with 15 rats in each group,which were given orally with ethyl acetate extract solution of hematoxylin 10 mL·kg^-1(2.35 g·kg^-1 crude drug quantity),cyclosporin A(CsA)10 mg·kg^-1 and the same amount of normal saline for 7 days.The donor heart beat was judged by abdominal palpation after operation,and the end point of rejection was stopping beating.The survival time of heart transplantation was observed,the morphological changes of myocardium was observed by hematoxylin-eosin(HE)staining,the apoptosis of myocardial cells was detected by TdT-mediated dUTP nick-end labeling(TUNEL),and the expression of caspase-3 protein in myocardium was detected by immunohistochemistry and Western blotting(WB).Results The average survival time of transplanted heart in test group,control group and model group were(29.53±3.14),(31.25±3.69),(8.65±1.28)d;the Stanford standard classification of rats were 2,2 and 4 grades.The apoptotic index(AI)of rat cardiac myocytes were 14.32±3.34,18.96±2.54 and 46.47±2.37,respectively.Immunohistochemical staining showed that the relative expression of caspase-3 protein in test group,control group and model group were(8.41±2.55)%,(10.52±2.34)%,(21.43±2.13)%;WB detection showed that the relative expression of caspase-3 protein in rat cardiac myocardium were 0.96±0.14,2.51±1.03,2.15±0.24;the difference were all statistically significant(all P<0.05).Conclusion Ethyl acetate extract of hematoxylin can inhibit cardiomyocyte apoptosis,alleviate myocardial injury and prolong the survival time of rats by reducing the expression of caspase-3 protein in myocardial tissue.
