摘  要:目的研究乌药对高脂血症模型金黄地鼠的作用及其机制。方法用高脂饲料饲养建立高脂血症金黄地鼠模型。根据血清总胆固醇(TC)水平将动物随机分为4组:正常组、模型组、依折麦布组和乌药组,每组10只。正常组与模型组给予蒸馏水灌胃,依折麦布组给予依折麦布10.0 mg·kg^-1灌胃,乌药组给予乌药有效部位1.0 g·kg^-1灌胃,1次/天。于给药第2,4周末采血,全自动血液流变仪检测血液流变学指标;以实时荧光定量-PCR法检测肠组织三磷酸腺苷结合盒转运体B1(ABCB1)和低密度脂蛋白受体相关蛋白(LRP)基因表达水平。结果正常组、模型组、依折麦布组和乌药组的高切还原粘度分别是11.62±2.03,15.37±2.57,12.47±1.10和11.60±1.66,模型组与正常组相比或者乌药组与模型组相比,差异均有统计学意义(P<0.01,P<0.05)。正常组、模型组和乌药组的ABCB1基因表达分别是1.88±0.32,7.27±0.77和9.06±1.47;这3组的LRP基因表达分别是1.96±0.27,1.77±0.35和2.68±0.32,乌药组与模型组相比,ABCB1和LRP的表达均增加,但差异无统计学意义(P>0.05)。结论乌药具有防治动脉粥样硬化的潜在作用,其作用机制可能通过调节胆固醇逆转运过程来实现的。Objective To observe the effect and underlying mechanism of Lindera aggregate(Sims)Kosterm.on golden hamster models of hyperlipidemia.Methods The golden hamster models of hyperlipidemia were established by feeding high-fat diet.According to the serum total cholesterol(TC)level,hamsters were randomly divided into 4 groups:normal group,model group,ezetimibe group and Wuyao group,with 10 in each group.Hamsters in normal group and model group were given distilled water by gavage.Hamsters in ezetimibe group were orally administered with ezetimibe(10.0 mg·kg^-1).Hamsters in Wuyao group were orally treated with the effective parts of Lindera aggregate(Sims)Kosterm.(1.0 g·kg^-1),1 time a day.Blood samples were collected at the end of the second and fourth week of administration.Hemorheology index was measured by automatic hemorheology instrument.The expressions of mRNA of ATP binding cassette transporter B1(ABCB1)and low-density lipoprotein-receptor-related protein(LRP)in intestinal tissues were detected by real-time fluorescent quantitative PCR.Results The numbers of high shear reduction viscosity in normal group,model group,ezetimibe group and Wuyao group were11.62±2.03,15.37±2.57,12.47±1.10 and 11.60±1.66,respectively.Comparing between model group and normal group or between Wuyao group and model group,the difference was significant(P<0.01,P<0.05).The expressions of ABCB1 in normal group,model group and Wuyao group were1.88±0.32,7.27±0.77 and9.06±1.47,respectively;the expressions of LRP in the three groups were 1.96±0.27,1.77±0.35 and2.68±0.32,respectively.Compared with the model group,the expressions of ABCB1 and LRP in Wuyao group were increased,but not significantly(P>0.05).Conclusion Lindera aggregate(Sims)Kosterm.has potential application in the prevention and treatment of atherosclerosis.The mechanism may be related to antagonize hyperlipidemia by regulating the reverse cholesterol transport process.
