摘  要:环状核糖核酸(circular ribonucleic acid,circular RNA或circRNA)是一类无5′帽子结构及3′poly A尾,以共价键连接形成闭合环形结构的非编码RNA分子,通过转录后调控的方式参与动植物细胞的众多代谢途径.莱茵衣藻(Chlamydomonas reinhardtii,C.reinhardtii)是一种重要的真核单细胞模式微藻,为探究其是否存在环状RNA,本研究通过RNA-Seq高通量测序分析正常与缺硫培养的C.reinhardtii RNA组学数据,首次预测获得218条环状RNA序列,通过对比缺硫处理前后莱茵衣藻环状RNA序列,获得8条差异表达的环状RNA序列(其中,5条为表达上调、3条为表达下调).本研究首次对单细胞真核微藻莱茵衣藻进行环状RNA的生物信息学预测,确认莱茵衣藻环状RNA响应缺硫胁迫,为下一步揭示莱茵衣藻环状RNA参与响应莱茵衣藻缺硫的作用机制奠定基础.Circular ribonucleic acid(circular RNA or circRNA)is a class of non-coding RNA without 5′cap-structure and 3′poly A tail,connected by covalent bonds to form a closed circular structure,and involved in numerous metabolic pathways in animal and plant cells through post-transcriptional regulations.Chlamydomonas reinhardtii(C.reinhardtii)is an important eukaryotic single-cell microalga.In this study,218 sequences associated with circRNA were predicted for the first time in C.reinhardtii.By comparing the sequences of circRNA from sulfur-replete and sulfur-deprived C.reinhardtii,8 differentially expressed circRNA sequences were obtained(5 of them were up-regulated and 3 were down-regulated).This is the first report on a circRNA bioinformatics prediction of C.reinhardtii,confirming that the circRNA of C.reinhardtii responds to sulfur deprived stress.
