摘 要:为研究谷氨酸(Glu)对脂多糖(LPS)刺激断奶仔猪肝脏组织及血细胞分类计数的影响,试验选用杜×长×大断奶仔猪24头[平均体重(7.02±0.21)kg],随机分为4组,分别为对照组、LPS组、LPS+1.0%Glu组、LPS+2.0%Glu组,试验期为28 d。在正式试验第28天,试验组腹腔注射100μg/kg体重的LPS,对照组注射等量生理盐水,4 h后采集血样并进行屠宰,取肝脏样品待测。结果表明:LPS刺激导致血细胞分类计数发生显著的变化(P <0.05),同时使肝脏组织中Toll样受体(TLRs)和核苷酸结合寡聚域受体(NOD)信号通路关键基因m RNA表达量显著上调(P <0.05);添加1%或2%Glu可显著缓解LPS刺激导致的白细胞、嗜中性粒细胞、嗜中性粒细胞比例、血小板指数的降低,增加了单核细胞比例(P <0.05);同时1%或2%的Glu也显著降低了肝脏组织NOD信号通路中NOD1、NOD2、受体相互作用蛋白激酶2(RIPK2)、肿瘤坏死因子-α(TNF-α)的m RNA表达量(P <0.05)。由此可见,LPS刺激激活了肝脏组织炎症信号通路,而添加Glu抑制了炎性细胞因子的产生,进而缓解LPS诱导的仔猪肝脏损伤。The purpose of this experiment was to study the effect of glutamic acid(Glu)on liver tissue and blood cell count of weaned piglets stimulated by lipopolysaccharide(LPS).Twenty-four Duroc×Large×White×Landrace weaned piglets[(7.02±0.21)kg initial body weight]were selected and randomly divided into 4 groups,which were respectively control group,LPS group,LPS+1.0%Glu group and LPS+2.0%Glu group.The experimental period was 28 d.On the 28 th day of the formal test,the test group received intraperitoneal injection of 100μg/kg body weight LPS,while the control group received the same amount of normal saline.Blood samples were collected and slaughtered 4 h later,and liver samples were taken for testing.The results showed that LPS stimulation resulted in significant changes in blood cell classification count and up-regulated mRNA expression of Toll-like receptor(TLRs)and nucleotide-binding oligodomain receptor(NOD)signaling pathway key genes in liver tissue(P<0.05).Addition of 1%or 2%Glu could significantly alleviate the decrease of leukocyte,neutrophil,neutrophil ratio and platelet index caused by LPS stimulation,increase the proportion of monocytes.At the same time,1%or 2%Glu also significantly reduced the mRNA expression of NOD1,NOD2,receptor interacting protein kinase 2(RIPK2)and tumor necrosis factor-α(TNF-α)in NOD signaling pathway of liver tissue.The results indicated that LPS stimulation activated the inflammatory signal pathway in liver tissue,while Glu addition inhibited the production of inflammatory cytokines,thus alleviating LPS-induced liver injury in piglets.
