吴茱萸不同提取部位致大鼠肝毒性研究

摘　　要：目的研究吴茱萸不同提取部位对大鼠肝脏的影响。方法选用SD大鼠为实验动物,依据临床剂量给予大鼠灌服吴茱萸不同提取部位药液,连续7 d给药,观察大鼠的活动情况、毛色变化以及体质量信息,给药7 d后经眼静脉取血,以血浆中ALT、AST水平变化,肝脏病理学检查结果来衡量大鼠肝损伤大小,进而评价吴茱萸的肝毒性。结果各实验组大鼠表现为腹泻、毛色暗淡、生长缓慢等。大鼠血浆生化指标结果得出,5个不同提取部位对大鼠肝毒性强度为:石油醚部位>正丁醇部位>水部位>氯仿部位>乙酸乙酯部位,病理学检查显示高剂量组的大鼠肝脏出现不同程度损伤。结论吴茱萸不同提取部位均具有肝毒性。Objective To study the effect of different extracts of Evodia Rutaecarpa on rat liver.Methods SD ratswere selected as experimental animals and administered with extracts from different parts of Evodia Rutaecarpa inaccordance with clinical doses for seven days.The activity,hair color changes and body weight information of therats were observed,and blood was collected through ophthalmic vein after 7 days of administration.The changes of plasma ALT and AST levels and the results of Liver histopathology were used to measure the magnitude of liver injuryin rats,so as to evaluate the hepatotoxicity of Evodia Rutaecarpa.Results The rats in all experimental groups showeddiarrhea,dull fur and slow growth,and the results of plasma biochemical indexes of rats showed that the toxicityintensity of 5 different extracts to rat liver was as follows:Petroleum ether extracts > n-butanol extracts > aqueousextracts > chloroform extracts > ethyl acetate extracts.Histopathological observation showed the different degreesdamage of liver in high dose group.Conclusion All extracts of Evodia Rutaecarpa showed the hepatotoxicity on rats.
